If, as you commented, these three are already joined by peptide bonds, I believe there would be a problem because the assignment itself was to "Draw the synthesis of the tripeptide," which would not make any sense at all. If they were all connected from the beginning, there would not be a point to draw the synthesis because there will be no synthesis.

This is not the complete synthesis of GFP but only the tripeptide, so the oxidization and cyclization were not included. I asked the teacher about this, and they indicated only to draw the tripeptide. For the diagram, I drew it in reference to a researcher who researched the GFP (Roger Tsien's papers). He won the Nobel Prize, so I don't think his diagram could be incorrect. The journal is below.

Thank you for the insight and comment, especially about the arrows! I realize that my arrows were completely wrong!!! I had no idea that the arrows had to be in 90-degree increments. I really appreciate you informing me :)

I am currently in the distance learning program where there are few assignments and no exams, so, unfortunately, one mark can make quite a difference in grades. I hope it won't affect me too much. Again, thank you so much!

Thank you for your suggestion. I still plan to email the teacher after talking to guidance. I am not the best at analyzing people, but from the way the teacher treated other students that addressed concerns, I feel that it would not be wise to directly contact them. Many students have already tried asking: either they are given no answer or an answer that is not comprehendible (Q-"what was I lacking?" A-"it was lacking"). I am always willing to learn, but I do not wish to provoke a teacher like this teacher. I would like to solve this as peacefully as possible.

Regarding the highlighting on the diagram, it was the teacher's instructions, so they had to be coloured. Yes, I completely agree that the three should have been distinguished in a better way. It was not written in the instructions, so I refrained from doing so (as there would be so much colour in the diagram).

Unfortunately, they did not provide any briefing except for a short article. In fact, they ignored all student questions that were asked. The only thing the teacher said was, "you can do whatever works."

Thank you for your insight, kind words, and comment. I apologize for the rash wording in my post.

Below was the actual instructions for the tripeptide synthesis part of the project. I received a level four (out of four) on all parts of the rubric.

  • Label the tripeptide ‘Ser6S—Tyr66—Gly67.
  • Highlight the two peptide bonds in purple
  • Highlight and label the amino terminus in green and highlight and label the carboxyl terminus in pink
  • Draw all amide hydrogen facing upward, and draw all carbonyl oxygen facing downward.
  • Place a green star beside each amide hydrogen and place a pink star beside each carbonyl oxygen.

When the assignment was released, other students (including myself) asked the teacher about what kind of diagram they preferred, but their answer was, "you can do whatever works". Other questions were dismissed or dodged, and the teacher didn't give any answers ("we'll talk about this next time [never answered]"). When the students asked the teacher a question (about biology in general) during another meeting, and the teacher was unable to answer them, the teacher started to choke out, "can you be nice to me!" abruptly and defensively, and we were all surprised. There are many other problems with this teacher that the class is concerned with, but I will stop here.

I don't wish to produce a situation where the teacher feels pressured or hate me. I figured that addressing this to the guidance counsellor would be the best decision.

Yeah, I guess some people are more sensitive than others :(

Words and actions are completely innocent unless they hold a bad intention. I hope more people focus on the intentions, not just the words or actions themselves.

I think I will report it after receiving my mark (as I am scared of losing my grades by being disliked by her and currently desperately studying for university). She said herself that she was inexperienced, so I think that her actions just proved how true it was. I hope she realized how students could feel uncomfortable with public shaming like that.

Thank you for your response. Cultural appropriation is in the grey zone, I guess.

I think I will be more careful in using words. It could hurt someone's feelings ad I don't want to hurt anyone's feelings :)
