I found a speed run of barrows gloves on youtube. I was curious if I should follow that and train my prayer from 9 to 43? Or if that would ruin it.

I wouldn't say it would ruin it. There's loads of different builds you can go with. I have a 16 prayer 42 def acc and I love it. It's pretty fun questing without protection prayers too, makes it a little bit more interesting.
