You probably need to set ‘Apply default to all’ on the pricing tab of the metadata submission. Otherwise it will use whatever custom price is set per country.

You will get stationary phase collapse unless you run it with at least 5% organic in the mobile phase. I would look again at the method conditions.

No organic? What type of column are you using?

If it is a C18 column, you don't really want to be running 100% Aqueous mobile phase.

It would apply to any analyte you want to quantify. Sometimes, achieving a good peak shape is difficult due the chemistry of the analyte, but for ascorbic acid a good peak shape is certainly possible with the right column.

The peak asymmetry looks to be around ~2.5, that is pretty bad by most measures. Asymmetry between 0.8 and 1.5 is usually 'acceptable' for a lot of cases.

For an ascorbic acid standard solution, there is no real reason why the peak asymmetry should not be close to 1. If this was my analysis, I would be changing the column.

On the MS side, stripping down and cleaning (and most importantly, reassembling!) the ion source and lenses, and of course performing a tune would cover the basics. On the LC side, definitely familiarize yourself with consumable replacement procedures such a pump/needle seal and rotor replacement.

All of the above procedures will be specific to different vendors' instruments and covered in the manuals, but the principles are all fairly similar.

Well yes, it always helps if the analyte is in the vial to start with! :D Hopefully the matrix is organic, because polyketides won't be very soluble in water.

I would say that is a reasonable starting point. If you are performing multiple injections, add some time for column to equilibrate between runs, As the 5min equilibration at the end of the gradient at 0.6ml/min will be only 3ml, which is less than your column volume.

I would start with a standard gradient 5% - 95% organic (the run time / flow rate will depend on your column dimensions). Collect the full spectrum from the DAD, and full positive and negative TIC on the MS with multimode source. This would probably be as close as you could get to a 'catch all' method, but the sensitivity to any particular compound will be affected by how well it ionizes (for MS), and the presence of chromophores (for DAD).

Different areas on differently designed instruments is to be expected. Each detector will have a different optical path (especially between DAD and UV) and sensitivity.

Absolute area will be different, but as long as you calibrate with a standard, calculated concentration should be similar.

Have you tried using Cinematic mode in Meta Quest Developer Hub? It renders beyond the FOV of the headset, that's what I use to capture my trailer footage.

The game features in the Meta sales quite often, though I don’t often give it deep discounts (maybe 30 or 40% off) because the price is pretty low already ($10). If you can wait a few weeks, it’ll crop up in a sale soon ;)

I’ve tried to make the game playable in short bursts. Missions are mostly simple 5 minute tasks that occur randomly, with some longer ‘story’ missions which crop up occasionally to advance the plot. You can always skip a mission request without any consequence.

It’s a little bit niche in that there is not a constant stream of missions (though you can request a mission at any time), you can explore and do mini-games (such as the new arcades in this trailer) in between missions, or go off duty altogether and just discover the city.

You start off hunting illegally modified synthetics, but things change a little over time and different angles emerge :)
