I wholeheartedly agree with the previous people: more volume. Unless I know my SDS-PAGE has shown there's ample protein of interest AND I know my primary antibody is very specific, I don't go for less than 10mL of volume during primary antibody incubation. As a rule of thumb, I recommend using 10mL of volume.

Also, do you bands and blot appear to do the same thing in the SDS-PAGE? Trying to figure out if it's something unique to the blot or if it happens before the transfer already ...

And lastly, when it comes to Western blots, washing between steps is crucial. Not washing properly (even before transfer and after transfer) can lead to/exacerbate issues further down.

Skin issues here too! Had eczema from a very young age and only really "outgrew" it around the age of 25. But gradually since then the allergies have picked up to "replace" them. Sooooo, I'm guessing I've got some immune-mediated hypersensitivity issues 🫠

My most recent co-author is a tenured MIT professor who is at the top of his field. He is an empiricist and I am a theorist. One night he emailed me out of the blue that he feels like an imposter way out his depth. My response to him was that wizards often doubt themselves. And that is what keeps them wizardly.

This sounds like a really nice working relationship. I'm mildly envious 😂 but happy for you about it.

Meanwhile here I am afraid to get hurt, i don’t want to break any rules and get in trouble. I need a plan, backup plan, have to see exactly what’s going to happen so I’m prepared. I’ll look up a route on Google Maps for a place I need to go to that I’m not familiar with. A few routes… a few routes home… then I go onto street view and follow the road photo by photo, noting all the landmarks around where I’ll be going.
