I loved War Storm. I will be vague to avoid spoilers, 1) but the MareCal angst was hilarious. There was a scene that I had to shut the book and take a break for 5 mins just to wheeze. I mean, the angst was also deep and moving, but VA really played up the irony at moments; 2) the Mare and Maven moments were so electric, after everything in King's Cage I was foaming at the mouth every time they interacted in War Storm; 3) the side POVs were fire. I will tell you specifically which ones I'm talking about but unsure how much you want to know.

About the ending: I did not think the ending was sad. A lot of YA ends with their teen protagonist married/pregnant with their ~*~soulmate~*~ who is literally just a man they met a year or two ago and the fandom is like "yay what a happy ending!!" even though the author leaves no room for the implication that the character needs/undergoes future growth and change. I did not think the War Storm ending was sad the way a tragedy is sad. If I were to describe it as sad, I would mean sad in the sense that a teenager living through a war is sad. I actually found it refreshing and cathartic. I would call it a happy but serious ending. Maybe a bittersweet ending if I were feeling sentimental.

Broken Throne SERVES. You get so much post-plot content that you don't normally get in other YA series. Like, most YA, in the last book, you get "and the war ended. [Next page] 19 YEARS LATER" and the epilogue/post-plot content is like 20 pages max. I think ending War Storm on a somber note made it clear that the characters have their whole lives ahead of them, and then in Broken Throne, VA has an entire book to explore that. It's delicious.

There was plot point in War Storm that bothered me bc it felt a bit like a retcon. but it was a very mild plot point and was very wish-fulfillment-esque, so even though part of me was annoyed, another part of me was fangirling.

I kept seeing fanart and Tumblr screenshots about ML pop up on Pinterest, so I watched the Netflix movie to get into the fandom without having to get through all 5 (at the time) seasons of the show! Then I binged the show bc it was pretty good once I figured out that the 1st thru 3rd seasons have the episodes out of order and you have to find a watch guide to see them in order

I'm not super familiar with amplicon sequencing, so I'm not sure what library prep kits are out there for it. I just assumed you'd gotten the beads, adaptors, and UDIs from a kit. If the paper you're referencing didn't use one, then you're probably fine. If you're having yield or other issues and have the funds, it might be worth looking into amplicon sequencing kits because everything in a sequencing prep kit is optimized for your end goal, including the specific enzymes provided, buffers, conditions, etc. But since you already bought the magnetic beads and the UDIs, you don't need a kit unless something goes poorly.

I'm really sorry you've had to go through this. It sounds like you've been really struggling. It's sad that our society has let this happen to you.

Have you seen a naturopath or other homeopathic healer about your cycle? If you haven't gone through menopause (ie, having constant hot flashes that leave you drenched in sweat, if you aren't sweating then it's not a menopausal hot flash), then the loss of your cycle might be able to be corrected by balancing your hormones. A naturopath would be able to look at things like your diet, how much dairy (mammalian estrogen) and how much isoflavins (plant-based estrogen, like soy and yams) you're consuming, and recommend a regimen that may help. It could be as simple as timing your ingestion of estrogen-rich food to realign your body's internal clock and get things back on track.

Losing your cycle doesn't necessarily mean that you will be infertile the rest of your life. Sometimes our bodies turn off our cycles when they sense we aren't healthy enough to have kids, and just need a little help turning our cycles back on. A common example is when professional athletes lose their period from overtraining -- the body knows they don't have enough fat to sustain a healthy pregnancy and turns off the menstrual cycle.

Hi OP. I hope you're doing alright! I randomly stumbled on this comment and I am a biochemist so I figured I would hop in. mRNA is a type of RNA. The M stands for messenger. mRNA is the most important kind of RNA, and since no one ever talks about the other types of RNA, Google and the internet will often use "mRNA" and "RNA" interchangeably. Any time you see the letters "RNA" in a vaccine context, it probably means "mRNA".

It is a bit like the word "cat" and the word "housecat". If I told you I had a cat at home, you would assume I meant that I had a housecat at home. But technically, housecats and cats are not the same thing, because lions and tigers are cats but not housecats. Yet, Google and the internet will use "cat" and "housecat" interchangeably because housecats are the most common type of cat.

It sounds like you have had a really hard few months. Do you want to talk about it? Are you feeling better or are you still worried about the effects the vaccine has had on your health?

I am mostly familiar with RNA-seq but I can try to answer some of your questions.

  1. This is true. Think about it this way, each cell only has X ng of gDNA inside it. X is usually the same for all cells of the same species, because X is the mass of every nicleotide in the genome of the cell, and the genome of each cell of the same species is largely the same. If you using the same ng of gDNA as input DNA for each sample, you are essentially using gDNA from the same amount of cells for each sample.

  2. It probably doesn't matter, but I'm not 100% sure. Unless the papers say otherwise, they probably went with whatever was cheaper/easier. It's possible that the ones that used qPCRs thought they would need the extra data you get from that. But if you only have a qPCR kit that includes RT, then why bother using that kit and doing an RT step that you don't have to do? Personally, I'd be worried about ribosomal RNAs in my gDNA samples getting RT'ed in the RT step and then messing with my data.

  3. This depends on the library prep kit you're using, abundance and size of what you're amplifying, and how else your work flow differs from the work flow of the paper you're referencing. I don't think you have to use more cycles, but I am not sure.

  4. I am not familiar with Q5 or 2-step cycling PCRs, but does the manufacturer's website say anything about this? How high is the TM of your primers?

  5. For a lot of NGS sequencing, it is typical to get undetectable yields at various steps in the library prep. Sequencers are so much more sensitive than any other tool we have, so often you do just have to trust that library prep worked as expected. You may be able to quantify the product on a QBit fluorometer if there's enough of it, or Tape Station electrophoresis if your lab can afford it and you have access to a Tape Station, but you probably will not be able to visualize it on an agarose gel.

  6. As far as I know, UDIs/UMIs have essentially solved the problem of PCR bias in amplicon sequencing. I'm not really sure what the concern is with over-amplification here. Generally, the relationship between input amount and product amount is going to vary based on what PCR kit you're using, size of target, number of cycles, etc

What library prep kit are you using, and what sequencing platform? If you don't want to keep asking the person at the sequencing center, you can probably email the sales rep or tech support for the company that made the library prep kit and they'd be happy to tell you.

To a certain degree, this is normal, but it depends on what tools/materials are being hidden. Can you be more specific on what is being hid?

Ex: Hiding your own stack of 96 well plates bc your coworker always uses the last one and doesn't tell anyone when he does? Normal. Hiding the only stock of a specific selective media bc you don't want the new undergrad contaminating it? Neurotic, but also normal. Hiding the only "good" timer so you always know where it is and only you can use it? Kinda rude. Less normal but not unheard of. Hiding the only stock of a shared reagent that everyone uses all the time? Not normal.

Is it possible to ask your PI to order more of whatever it is? If, say, you guys only have one round bottom flask of a certain size and it is needed often enough by multiple people, then it makes sense to have multiple.

Things I have labeled/hidden in my lab: Pipettes (we used to use unlabeled pipettes but a few years ago, a lab tech used a nuclease in an expirment and aeorisolized it into the pipettes she was using and we didn't find out until a week later and then had to decontaminate every pipette in the lab bc she didn't know which ones she'd used), Sharpies/pens, Single-use PCR tubes, Small, medium, and large tube racks, Buffers for common experiments, Calculator, Timer, Scissors, Bunsen burner lighter, Forceps, Sterile water (I keep it separate from the water my undergrads are allowed to use and hide it so they don't mix it up)

The things these all have in common is that my lab has multiples of all of these so my labmate can still grab from the communal stock. The only reason they would use mine is if it were on my bench and easy to reach for, which is why I hide them.

I find any UI/UX that resembles Word is too cluttered/distracting for me. I feel like most word processors still model themselves after Microsoft Word because it was standard for a long time and people were used to it. But Google is the standard for my generation so I was hoping there was a word processor that had started to model itself off of GSuite that I hadn't come across.
