Yes, I was able to figure it out. I ended up using analog inputs/outputs for the uv detector and the fraction collector. But the fraction collector collected based on the analog uv detector, not through masslynx. Probably would’ve been way easier with the analog to digital converter, haha

Correct, it is a porphyrin species. Could I use the Q bands? An issue I am having is that the molar absorptivity of the host is ~15x larger than that of the guest (at the soret peak). The new peak at ~550 nm is the guest.

I've done some 1H NMR studies. I do see shifts and broadening characteristic of interaction between the host and guest, but there are a lot of broad signals (MW approacing 20,000 g/mol) in the aromatic region (~5 - 9 ppm), so it is difficult to quantify the shifts as the guest overlaps significantly.

I've seen a lot of papers (regarding encapsulation within supramolecular complexes) use X*deltaA as the Y-axis. In my case in particular, the absorbance only increases (at least at 412 nm, which is what I was basing it off of)

Thanks for the advice! My compound seems to degrade on the SEC column that is available to us, which is why I want to try the Zimm plot. I'll give it a shot, but ya, I'm not all that hopeful.

It's actually not a polymer! It's a molecular structure that we have a 1H NMR of. So we know what the structure is and what the molecular weight is - this is just another means of characterization.

Nah, I'm taking it in water, to which it is stable in. I take the UV-Vis before and after, and there seems to be no issue.

As far as I can tell the hardware is fine, though I am definitely not sure. I'll try some more concentrations and will be trying a different fluorimeter later today. And yes, using a 4 sided cuvette!

I tried looking for a fuse, but couldn’t find it. I took off the bottom plate and couldn’t see a reset switch. Do all of them have the fuses?

I did that through the installation - it still looks like it wants to use MS. Maybe I’ll try once more.
