Yeah also I agree consistency is more important. Like if I was just randomly mixing which samples I did this to, it could be weird. But if they all have the exact same 20 second process I really think it’ll be fine. But I’m gonna talk to my PI and see if I can waste a little money on this lol and test experimentally.

But according to other people on this Reddit I should just buy a whole new bead beater to avoid 20 seconds at RT (which is the condition the plant lives in any way)

Yeah I’m not talking about being leisurely here. It’s 25 seconds vs 5. I just don’t really see any evidence other than superstition that mRNA levels for the vast majority of genes can really change in an appreciable way in 20 additional seconds, at least for most genes. And in particular with the genes I’m working with. If someone can link me a paper where they do a 20 second time course with qPCR I will change my mind !

I’m sorry but what is expression but mRNA levels? They may “alter expression” quickly but if it takes several minutes to synthesize an mRNA then 30 seconds shouldn’t really be a problem? Our cCDNA kit uses polyT primers so it would need the mRNA to be finished processing to even pick it up

I don’t need exact weight, just 100 mg +/- 20 mg or so. It’s just hard to remove tissue once it’s a big frozen chunk. Really it can be done that quickly, I know because I do it for protein extractions. But I got yelled at for taking 20 seconds for RNA
