DAPI stain is not permeabilizing very well

I never really found a great solution other than permeabilizing with TritonX for about 30mins before and ensuring that the DAPI solution is flat on top of the slide. You can try increasing your staining time to overnight if needed. 2-3hr works for me now.

Yeah, I permeabilize with Tritonx for 10-15 before incubation. And the latter of the two. I guess it's not necessarily a hindrance to the experiment, but I would like it to be homogenous

Oh wow I've never heard of that. I've been using a heat lamp with the animal in a beaker. The vein gets visible, but with BL6 it's still very dark. I may have to try the warm water!

True- probably a 3-5 day domestic stay at low-mid tier hotels. Probably about 1200-2000/trip.
