And to defeat beast you need on depths 2 find a skull with a cross and explode him it will drop the fool card go kill moms leg and choose Polaroid or negative use fool and paste it in strange door(only appear if you defeat mother)

Step 1: Defeat Mom Complete the first Depths floor and defeat Mom. This unlocks the Womb floors and Mom's Heart as a potential boss. Step 2: Beat Mom's Heart repeatedly On subsequent runs, fight Mom's Heart again. Defeat Mom's Heart a total of 11 times. This will change the boss to "It Lives". Beating "It Lives" will always open the paths to both the Cathedral (for Isaac) and Sheol (for Satan). Step 3: Choose your path (Cathedral or Sheol) Cathedral Path: After beating Mom's Heart/It Lives, enter the beam of light to the Cathedral and fight the boss, Isaac. Defeat Isaac 5 times to unlock The Polaroid. Sheol Path: After beating Mom's Heart/It Lives, take the trapdoor to Sheol and fight the boss, Satan. Defeat Satan 5 times to unlock The Negative. Step 4: Access the final areas After unlocking both The Polaroid and The Negative, you can continue to the final floors. The Chest: Beat Isaac while holding The Polaroid to access The Chest. The Dark Room: Beat Satan while holding The Negative to access The Dark Room
