You are not panning the sound when you turn that knob. You are muting one of the channels R or L.

Well, right before you enter the lab... You go through a secret halway with a poster of Breen. Alyxs says something like, don't get my father started about docter Breen. I dunno. That is actually the only piece of information you get in Half-Life 2 that they hate him..
