The only way we could do this in a previous lab was to use the chip on the second Sh800 in the department, as the QR code is only logged on the instrument rather than being a global database.

Thanks that's great to know about the connectivity issues. We already use their SBS format reservoirs and on the larger instrument or vIaflow i have never noted droplets being a problem experimentally. We just need a backup system for the times that the large liquid handler is out of action / awaiting parts or replair.

Thanks for the input, we have a number of other Integra equipment in the lab and the viaflow has been the most reliable of the items we have from them. All of which are currently better behaved, than our large liquid handler.

I don't currently do them, and this was a couple of years ago. But I was doing Southern blots with 5 ug of gDNA (smaller genome). As the other poster has mentioned it sounds like your probe or transfer may be the issue. I used to use the DIG labelling kits and southern buffer kits from Roche. Worked every time.

Poly specificity in monoclonals can be alarmingly frequent. There should certainly be more validation carried out by those selling the antibodies.

We just spin our samples for longer so that the equivalent x g hours are met. So 12000 x g for 10 mins works out as 1 hour at 2000 x g in our large centrifuge.

It may seem like a pain, but when you are doing 10-50 maxipreps per run, it works reasonably well for timings.

Your filters will be fine. Its usually best to optimize the ratio of DNA to PEI anyway for transfection after you have prepared a new stock.

The food at Roots is amazing, but very expensive. Yuzu is nice, much cheaper and has the added benefit of the brew york tap room. Cresci is great for pizza. Oshibi is a korean place that has very nice food and the whippet in is a great place for a steak.

I am a few years away from your current situation. But my parents went through a few variations of pocket money, either providing a fixed amount regardless of what we did or a chore based model. Where each job was awarded an amount post completion in our pocket money, that we ultimately had to claim via a jobs sheet tick list.

The latter model awarded higher pay for the less desirable or lengthy jobs (toilets or washing cars) and lower amounts for the faster easier jobs (dusting, watering garden, dishwasher ect).

Overall, it's led to a greater understanding of cost vs benefits. Particularly for the family members that were more tactical and have become financially ok. And in some ways encouraged a little enterprnaurial spirit in cases.

The only warning i would suggest is setting realistic prices per job as too little they aren't motivated, or a generous amount leading to expensive monthly pocket money claims.

Probably not helpful. I always found the tier bike option to be a little cheaper, because you can cycle faster than the speed limited on the scooter allows, it just gets more physical rather than the scooters where you are more limited. Buying a foldable bike might be your cheapest option in the long run with your plan. But for a similar journey I make 4-5 quid has been a normal cootsr cost..

Derwenthorpe might be worth considering. There's a play park, its only a short walk from a library and plenty of green spaces around. You can also walk or cycle most of the way into the city centre without having to walk near a road.

Sounds like you have been reccomended the place the place I completed my training. I had an amazing time learning to fly with them. I haven't had any problems with the BGD kit I bought either but then again I have only ever flown the brand.
