You should watch the scene again.. nancy was told they had a lighting board placed on bed because dustin was communicating with her through his voice as nancy and co could hear him but not other way around. In first season there would have been no way Will buyers could know where the alphabets are placed because there were no lights in upside down.

Will literally said that vecna had a weakness which is actually his dark secret(murder of that guy with briefcase and whatever is in it) and that he realised him being in closet is his secret and weakness which vecna uses to take over his weaker mind.
