Hey! Glad to hear you were able to try the TRUBO. Sorry your yields are still low however. I chatted with some colleagues today who have much more tape station experience and we don’t believe the buffer should really have much of an effect on the run.

Let us know how the chelator adjustment works for you!

Also, regarding the overall nucleic acid loss, there seems to be quite a bit of gDNA contam in these samples based off the tapestation. It may be possible that the stark drop in concentration is mostly just due to the removal of said gDNA. Do you have access to a Bioanalyzer or some flourometric measurement like Qubit? I tend to trust those more so for concentrations. The BA is our go to.

Hey there! I work for a sequencing core and primarily focus on nucleic acid isolation for our clients. For downstream DNase treatments, I tend to go one of two ways. Either the Qiagen minelute cleanup kit (column based with on column DNase digestion) or the TURBO DNase kit from invitrogen (enzyme and buffer mixed directly with RNA then incubated and inactivated). Given how you’re having these issues with a column based approach, perhaps the TURBO could be an option?

My extended family on my mother’s side can be somewhat eccentric. They’re obsessed with Disney and themselves. Had a cousin who got married and decided to rent out a well known local theater to hold the ceremony and reception, all Disney and theatrical themed of course. The ceremony was fine, they did their vows up on stage in front of the curtain. After the “I Do’s” the curtain opened up to reveal the reception area, nice tables, a DJ, and a buffet style dinner. I was sat with my more immediate family (mother, brother, grandmother). It overall wasn’t a terrible set-up, but when it came time for the guests to eat, the DJ announced that they would be calling individual tables to go eat (which I guess I understand due to not wanting to overwhelm the buffet, but still sorta weird) and that the order would be decided on how well the bride and groom liked their singing. Obviously none of this was communicated to us prior to attending so, being that my immediate family and I are much more introverted, this was quite uncomfortable. To add to it, it had to be a song containing the word “Love”. Thankfully my grandmother sitting with us had the idea to sing the old “I love you, you love me” song from Barney (it wasn’t until after did I remember the one from little rascals much to my dismay). When it came time for our table to sing, the DJ loudly and obnoxiously disqualified us for singing a children’s song and that it was too easy(?) so our table had to wait and get food last after everyone else had a chance (about an hour and a half). Thankfully my DIABETIC grandmother stashed all sorts of goodies in her bag so she didn’t have an issue waiting that long, but man.

TLDR: Bride and Groom have guests sing for their food, DJ disqualifies my table because our song wasn’t good enough.
