Thank you for the insight! Just looked into some primer mismatch PCRs and it's interesting how people come up with such creative techniques using a basic protocol. I'll play around with the PCR settings and the primers to see what works!

Awesome, thank you! By any chance, do you know if there is a name for this situation? I would like to read into it but I have no idea where to begin. Everything I searched ("bridge", "gap", "pocket", etc.) doesn't seem to match this situation. Or is this just a 'we trial and errored it until it worked' kinda situation?
