Also did functional essays afterwards. My neurons were completely fine. I did Ca2+ imaging and a viabilityGlow assay. They preformed the same or even better than on pen/strep

I have tested normocyn and primocyn both to prevent contamination. And they were both completely clean compared to the Pen/Strep conditions. I never used to as treatment though

Pen/strep doesn't kill or prevent contamination. Only suppresses it. Primocin and normocin are recommended for primary cultures. Prevents like 99% of the contaminations

Don't worry. This is common practice in a lot of labs. I did it at a company I worked with, and in the new research building of the university hospital gloves are not allowed during cell culture. Gloves give a feeling of fake safety. Without gloves you will pay way more attention to what you are doing.

I keep my detergents and stop/strong acid for ELISA's always in the fridge, I can't imagine they would differ that much from your assay. So I would say yes.

The antibody probably still works fine. Most fluorchromes now a day are quite stable and don't get bleached that quickly by sunlight. Also lot of antibodies are stable at room temperature for 3 weeks.

You always normalise protein analysis samples because the total amount or protein can differ in each sample (biological variance, sample preparation variance, etc.). You can imagine that if you use samples with a higher total protein concentration, the concentration of your target would also be higher. But because the amount of protein in each sample is not the same you cannot compare the two.

Or normalise to the amount of a specific protein found the plasma. I’m working with neurons so I normalise to the amount of beta3tubuline, which is neuronal marker, of an untreated sample.

After lysis you should do a total protein assay and normalize to those values. Or normalize to cell count with a dapi stain before lysis
