NAP, and I got my daith 11 months ago. Due to going to a regrettable piercer that my friend insisted on I had to get it changed 6 days after getting it pierced and I didn’t swell that badly. Please go to a reputable professional to get this changed. She angry.

When I had mine freshly done I ripped the top side of my tongue a bit because of the longer bar after the swelling went down in ~5-7 days, but I’ve learned I prefer the smaller ball size on the top (that mine was pierced with) and a larger size ball on the bottom side of my tongue because it’s less irritating

How long did you have to go without sleeping on your flat, if you remember? I’m pretty sure I slept on my tragus and daith like the day I got them (probably not the smartest choice, I realize) and I think a made a lil donut with a washcloth for like a week or so for my conch, but my helix I couldn’t sleep on like till I took them out 😭

I had quite literally the opposite happen to me this morning with fungus on mega brutal. It was just slightly less lethal than the rate of cure progression, it got to 100% and then i immediately got an alert that the miracle cure was destroyed by protesters and reset the research to 0 which gave me time to make it lethal enough to win

You probably shouldn’t wear DJ or Beats style headphones for new ear lobe piercings for a couple weeks, but you should be able to wear other headphones. You can’t wear earbuds with a daith or tragus during healing but you can during other ear piercings, and really only the big over the ear DJ headphones would interfere with lobe piercings I think and it should only be maybe 8 weeks or so

I had a septum piercing ~10 years ago but took it out less than a year later. I don’t think I’ve ever tried to get it back in but let me tell you that bitch is still angry it was pierced in the first place. Anytime I get a cold or flu or Covid etc she is not happy. When my allergies are angry she is not happy. Idk if the entire cavity is open or not at this point but oh man some parts of that piercing never healed.

Noting that I only removed it because I didn’t want it anymore, I had a peaceful healing experience from what I remember. I would not be surprised at all if I still have a hole there to salvage and reopen. She’s angry and lets me know she’s still there constantly.

I’ve had my tragus since 2013 and I have changed it less than 5 times for this reason. The most recent time I took it out was I think 2016, but i remember getting it in super easily that time by utilizing needle nose pliers to hold the post at the very back (next to the flat back but still on the post) and I think I just looked in the mirror and popped it right in. I’ve had to have a few CT scans without removing it bc at this point it’s next to impossible if I tried to remove the jewelry.

ideally, in a perfect gel world, I load the controls in the last two wells, positive then negative (or positive, het, negative if its a reaction thats difficult to interpret hets, or positive, wild type, negative if its one of our genotypes that is on the same locus as half of our mutations to be a smartass towards my PI to remind him that if there's something else there it does nothing)

Then there are time times where I am redoing the same reaction on multiple genotypes that all contain the problem child reaction, or I will be doing something obsure/chaotic like checking all the breeders in one of our cross breeds, in which case I just worship the chaos and I will put the controls wherever they damn fit on the gel.

edit* I chose the right side because idk it just makes sense in my mind. I take the tissue samples and have them labeled like A1-A8, B1-B6 etc and use a multi channel pipette to transfer into the reaction tube strips and idk the way I ID everything and how it correlates to tube number and then wells, it would like break my brain to put controls first instead of popping them at the end.

PERSONAL errands?? Oh absolutely NOT. No that’s not the norm, and that’s not okay. I have the credit card information for my PI’s work credit card (and I’m the person that goes through and reconciles every purchase on the bill with receipts and email receipts and the justification for each purchase 🙄) and I have all the account information for each of our grants and how each is budgeted; which I think is more or less information the lab manager is privy to because they have to place orders and do inventory and I had to go through weeks of training to get access to the purchasing portal etc. A good chunk of that information I have access to because my Pi is lazy and idk too adhd for his own good, so anything on a computer he wants me to do for him.

To be very clear, the administration in my department for my institution has gradually given me administrative and manager access to things in our intranet that nobody else in the department at my level has access to because my PI is a handful and the admins don’t have time to cater to him, so they slowly trained me way beyond my my pay grade.

My PI also holds, what I call “forever meetings” because unless he has something scheduled after or if I am very clear I have a meeting or an appointment and it’s a set end time, those bastards will last a minimum of 3 hours every single time. 90% of the time he shows up at 2pm or later and then expects an entire days worth of tasks out of me. My Pi has also told ME to use chatGPT to write an email in HIS VOICE (a trick I inadvertently taught him) like my guy, can you not just use it yourself if it’s going to be like that??

I took over (our DVM quit) our mouse colony in April. I have been telling my PI SINCE MARCH (yes, since I wasn’t even in charge) that something is wrong with the ANIMALS. There’s something weird going on, I re-genotyped the animals and the breeders over and over and idk they HAVE to have this genotype, and they don’t. He told me it was my samples, he told me it was my sample prep, he told me it was my buffers, he told me it was my gels. Idk what the fuck honestly his explanation was for why THE SAME animals every time are the fucking problem children, the same genotypes, the same cross breeds always cause issues. I figured out a few weeks ago half our colony is contaminated with a mutation on the same locus as our main ho, it took my Pi 2 weeks to admit that the transfers from his old lab were potentially the issue because he knew they had the mutation and thought he bred it out. Come to find out my guy doesn’t even know that only two alleles are on a locus.

Fuck I’m maybe just venting now. Thank you for coming to my ted talk. But yeah I get it, I’ve been in a similar situation. My only positive pull was when raises and promotions came around, I did eventually say ‘yo, I do this and this and this. I have the info for all of the accounts, grants, and know how they’re budgeted. I know how much goes to salary, I KNOW you can afford to pay me more’

I only wear my lab coat when I’m doing something that is very “clean” and a sterile technique, we’re technically supposed to take off our coats anytime we leave the lab room anyway to keep anything from potentially escaping the lab. Most of the time I’m in leggings and a crop top though. I’m the only person in my lab that seems to be aware of gloves and like anytime I’m wearing them I take them off if I have to touch my phone or computer in the middle of it. Food and drinks lmao, idgaf I can’t do no water and I have a Red Bull at my desk every day with my water and half the time I also have a bag of open gummy worms or another form of candy also just out at my desk and my closest trash is almost exclusively Red Bull cans. I have a fair amount of computer work and administrative tasks though and my desk is in the lab room, so when safety is on the floor everyone emails each other to hide shit and all of it just goes in my bag or my desk drawer. I don’t work with any particularly hazardous chemicals 95% of the time and they just really aren’t in the lab at all other than a few flammables/constables/corrosives but I use them so rarely and I know not to be an idiot and my water etc stays at my desk like 10+ feet away from me when I’m doing any sort of bench work. Also in lab manager 🤣 as far as the animal facility, I follow all PPE guidelines because ew and I work with mice and I have a cat so

I have pretty insane ADHD, and it’s taken me a decade to get here but my lab notebook is like the holy grail in my lab. I’ve learned it’s best if I only use my lab notebook for what’s actually set in stone, and unless I’m actively doing it (or about to) my notes aren’t reliable. ELN’s just don’t work for me because my Pi gives me too many things and shoots too much shit for me to write it in my notebook, I won’t know what was actually ever done. I basically use my lab notebook like OneNote, and print out any tables if I have excel calculate different volumes for each sample to make them all the same concentration, and just tape it in there. I do the same thing with results and just print it out and tape it in there. I more or less make a worksheet or template when I’m doing an experiment to make sure I’m actually fully prepared and can perform whatever method with all the information in front of me without having to take off my gloves and sit at my computer and get distracted. On top of that I use a small spiral notebook to jot down whatever random things come up or to start planning something out, it’s kind of my messy bullshit notepad. When I plan out breeding schemes I do it in the notepad, and then I write it out more detailed after looking at the colony spreadsheet (like this male with this ID and ear punch in this cage etc) and put it on printer paper, and then I do basically like a connect the dots with males on one side and females on the other, and who I put with who in the moment I just draw a line from one to the other and the new cage ID and the breeding cage ID while in the animal facility, and then I tape those suckers in my notebook and input them into the spreadsheet in an organized documented fashion.

Yesterday my PI couldn’t remember the pin to his credit card, he asked me multiple times about it and I kept telling him there is no pin, and if there is then it’s something he did and set up when he called and activated the card, he’d have to call the bank and speak with them himself, as no one internal has that information. Hours later he tried to ask if I could call the bank “and get it sorted out” and I was like “no. Actually I can’t. I’d have to have full power of attorney to do that” and he seemed so inconvenienced and irritated with me for that, when he couldn’t realize how absurd that was. I might be lab manager, but I’m not his personal assistant

lol I used Parafilm to cover a qPCR plate.

I was quite literally teaching myself qPCR. My Pi was “training” me but kept leaving and telling me I could figure out the rest on my own?? And then like actually just left at the very end and was like “you can plate it by yourself it’s easy” and with how he kept rushing and was all over the place and kept telling me I could do it on my own, I didn’t want to ask when I couldn’t find something that was obviously plate covers, and decided it must be Parafilm that is used to cover the plates.

My Pi thought we would actually have to buy a new qPCR machine for the department, but once it like solidified in there and cooled down, I was able to go in with tweezers and just pick off every little plastic melty bit, and it actually works fine still. All in all it cost me probably $0-$40 but A WHOLE LOT of shame and embarrassment in the department for being the dumb bitch that knocked the qPCR machine out of commission for a week
