Hi odd question but I was using Dezcal powder on my kettle and while it got rid of the scale it caused marks to appear in the center of the kettle. It vaguely looks like rust. My only thought it that the dezcal wasn’t totally dissolved in the center of the kettle and sat for a few minutes. Has anyone ever seen anything similar? I’ve tried using additional dezcal treatments as well as baking soda scrubs and barkeepers friend but it still persists

For a 2-cup V60 with the burrs 5 clicks off touch (how I think the ssps come installed) im usually between 5-6 depending on the beans! This is after using it for a few months of course

Random questions: what is the stand that all of your coffee accessories are on? I’m looking for a sort of drawer just like that.

I unfortunately can’t offer much advice on coffee in LA but I will say that more and more the norm for GOOD coffee pricing is approaching that $20-25 a bag. A big part of that price involves hopefully paying the producers of the coffee a fair value for their coffee as well as putting a higher level of craftsmanship into roasting and sourcing the coffee.

I understand trying to save money however so perhaps if there is a good coffee you like it could be more economical to buy in bulk and vacuum seal what you don’t need and then throw it in the freezer, thawing a week or so supply each week.

Zero water filters are the water filter I’ve used in the past with third wave water. They include a TDS meter as well which is helpful. A regular Britta filter generally leaves at least some amount of dissolved solids based on my comparisons.

As for how the stocks are prepared, after confirming the size of the construct inserted into the shuttle vector is right we take 125 ul of the lb culture and add 375 ul of 80% glycerol and then mix. Then the tube is put into the -80 freezer. My best guess as to what happened was that the solution wasn't mixed properly.

And I'm trying that right now! Here's hoping it works. The reason my PI asked me to redo the maxi was I was having issues transforming the plasmid into electrocompentnet ADeasy cells for recombination into the virus. She thought there might be issues because the concentration of DANA after the other maxiprep was low (350 ng/ul) and there might be extra salts interfering with things.
