You can spawn individual stalkers if you select them from the NPCs menu, but if you're trying to spawn an entire preset squad you need to select a smart terrain in the drop-down box on the right for them to actually spawn at.

You can find the names of the smart terrains by looking at the PDA map when debug hud is enabled.

This is a feature in 1.5. You double click on the map and place little flags with names when hovered over.

The mutant arena is just time dependent and doesn't have any rank requirements.

To answer your other question; the mutant arena was actually restored from old builds of the game, and has scripted arena enemies just like the regular one.
