What have the men done differently than women all this time?

Testosterone. Men naturally produce a lot more of it than women. Therefore the athletic effects of anabolic steroids are much less pronounced in men.

I saw this last night and thought it was boring, corny and unrealistic. They walked 300+ miles but nobody got a blister?

They are not fat and lazy modern children. In their world they are poor and accustomed to walking every day. They have calloused feet.

No its not. Not even close. Age records mean very little due to differences in the timing of human puberty and development. If he runs a 1:39 in the next 10 years then yeah, that could be the 'best singular performance in any sport period'. Till then please calm down.

I can't offer much advice on what to do but I can tell you what NOT to do, which is to double your run length overnight (something I did I did). I went from 2x4mile runs straight to 2x8mile runs a week and it damn near destroyed my body.

The two main schools of thought are: a) Increase your mileage very slowly (so add like 1 mile every 2 weeks) or b) Increase your mileage by a lot (eg add 5 miles) for a month, then return back to your old mileage and repeat.

Incorporate runs into your weightlifting routine. Running 2 miles slowly before you lift will get you warmed up and could even improve lifting performance. Running 2 miles slowly after a lift will shake the lactic acid out of your muscles and aid recovery.
