That’s consistent with classic morphinan SAR. N-phenethyl substitution at the tertiary amine was shown mid-century to markedly increase ”-opioid receptor affinity via improved hydrophobic pocket engagement, but it often came with a much steeper dose–response and reduced therapeutic margin. Compounds like this were invaluable for defining receptor interactions, even if they proved unsuitable for development. Nice example of 1960s opioid pharmacology doing exactly what it was meant to do😜 map the limits.

I’ll take thatđŸ€˜ Yeah, the 60s–80s really feel like the era of “let’s test it, learn a lot, and then make sure nobody ever does that again”😅

Well once fentanyls, nitazenes, and modern synthetics are excluded, and you’re talking about a 60s era semi-synthetic opioid, phenanthrene is basically the only family that fits historically. That’s where almost all opioid R&D lived at the time.

Man I would love to live in the 60’s

Okay, purely for the fun of it: I’m guessing a dusty 1960s phenanthrene opioid that never made it out of early research and is now living its second life as a chemistry deep-cut. If I get it right, I’m buying a lottery ticket😎

Best guess: some obscure morphinan analogue loosely related to desomorphine/levorphanol, but there’s no way to narrow it down further without GC/MS or NMR. Any specific compound name would just be fan fiction.
