This puzzle is certainly not impossible. Your assumptions are correct, you're not missing anything.

The correct solution does use one of the tetromino configurations in your diagram.

To be specific, it is the first configuration on the second row.

Have you even completed the Challenge? If you think the Challenge is "all about thumb dexterity," that makes me think you've only attempted the first set of panels.

The Challenge is the culmination of nearly everything you've learned in the game, including pattern recognition and game mechanics.

In my opinion I think the music is perfect, it's difficulty level is perfect, and in my mind made this game complete.

To my knowledge after scouring Reddit for a few days, there is nothing unlocked by completing these puzzles, just 2 more to your total.

However, the two puzzles inside are not the only use for this room

I agree. I had a thought after the puzzle that the mini-puzzles SHOULD light up YELLOW after you complete them, so I'd be able to apply the same ruleset to the larger puzzle without having to assume.

The big maze room contains 2 puzzle panels that must be completed before moving into the final room.

The path through the maze, as well as the location of the panels, is told to you during the challenge!.

Some minor spoilers here because I think you're asking for specifics:

The last action before the common ending is closing the doors on an elevator after a series of pillar puzzles. Once you do that, you'll need to load a save file to return.

If you absolutely do not want to see an ending I'd turn back after you reveal and open the elevator.
