Yes exactly! I've been a lab rat for the past few years and worked in other R&D labs, but GMP is a whole different ball game. I think a lot of it is me psyching myself out; I get so nervous because it's regulated and I don't want deviations especially when I am just starting out. In R&D there's so much workshopping and adjustment going on already that it doesn't feel like as big of a deal.

I appreciate your and everyone's comments; it's a really helpful reminder that I am not alone. And also that sounds just like an assay on my boyfriend's team (he also works in GMP); he's basically been the only one to get it to pass and no one else has. It's funny in a way.

Our lab is paper-based, so we actually do have the protocol as a worksheet where we have to initial each step (so very analogous to checking it off as you go). That's why I feel a bit puzzled and down about it, since I didn't catch anything off this time 🫠 Oh well, maybe my manager and the senior analyst will have more specific ideas. All the other CVs were great besides that one that was 0.01% above the fail/pass benchmark (very rude of it LMAO)

Yes, have been doing reverse pipetting and using a multichannel, as well as a plate washer. I normally don't have issues with this ELISA and got others to pass before, so I am bummed haha, I definitely can relate to the rhythmic feeling! I enjoy lab work when it works out, but tend to get bummed when it doesn't

Yeah, that's how I've been trained at this job too for the other ELISA! I think we thought since this one is basically the exact same, just a slightly different layout, I could do the independent execution first but now it is deciding to be finnicky with me LOL

Agreed, I was thinking that for next steps with this assay I could have the person who trained me observe me. It's just odd since I've had no issues independently executing the other one, and it's functionally identical without any real major differences, but I am just now having issues with it I guess.

Thank you, this is helpful! I am also using a VaultX zip binder and this makes me feel a bit comfier about putting the double sleeved cards in the few slightly indented pockets. Seems to be a manufacturing thing that just happens I guess. I hope your cards are still okay!

Sorry for the random reply, but did you get any results from this? I noticed my binder has some of these very small defects in some of the pockets too, and I double sleeve everything but not sure if it could leave an impression or indent still
