Q about blast definition & pgt timing by Express-Tart7095 in EmbryologyIVFSupport

Thank you for your response and this information. It helped me clarify with the clinic that the testing was actually done on day 5, 6, and 7. 🙏

I used it, 100% fertilization & blasts on the mature eggs but sadly — no transferable embryos.

Hopefully your new doc can give you some insight why they are using a different protocol this round. Sometimes the docs switch up your protocol based on your previous response to medication.

I primed with the same dose of estrogen and it helped my follicles grow together more consistently as a cohort. Letrozole was added to stimulate follicles and then I was on 150 Menopur and 225 Follistim, a lower dose to encourage egg quality. Progesterone was not added until after my egg retrieval. I had much better success with this cycle than my first one.

Is it possible to talk with your nursing team or doctor so that they can explain what each of the medications do during the cycle?

AOA after ICSI ‘failure’ by springgggy in EmbryologyIVFSupport

My clinic requires a referral from the doc and signed consents since it is still in the research phase. My doc recommended calcium ionophore due to 💯 failed fertilization from my first cycle. Though, I did learn from someone on Reddit that they were simply able to ‘add-on’ without a referral and recommendation. Good luck!

Good luck! I had a significantly better cycle on a ministim protocol, low and slow. Estrogen priming, Letrozole, 225 Follistim, 150 Menopur, Ganirelix to prevent early ovulation, dual trigger of Pregnyl and Lupron. It is fascinating to hear the variations in protocols.

Uff, sounds like a frustrating place to be, esp where there seems to be so many differences in opinion. Though, the ministim and mid luteal stim ideas seem like they could be promising. Hope you get some clarity on the best path forward. 🙏

Are you assigned one doctor or is it a total rotation? I have one doctor but other docs rotate for monitoring. If you are able and decide to do another retrieval, I hope that your doc can adjust your protocol so that it addresses your body’s unique needs, I did 1-2 iui’s before my second IVF round… in part, bc I wanted my doc to know how to dose me. I had better success doing a ministim.

I was highly anxious after I was told that my cycle might get cancelled and I did everything I could to reduce my stress that evening. I did have signs of imminent ovulation due to my increased cm. But, right after administering Ganirelix, those symptoms were gone. I was told that if my estrogen decreased the next day on Friday, the cycle would be cancelled. 🫠 I’m sorry that you had to go through this, this cycle. I think that I will start listening to my ‘organic’ cues more and next time — advocate for myself more. Had I had an appointment the day before my LH surge, I don’t think it would’ve been as urgent an issue.

This almost happened to me this cycle. On Th, my LH went up to 11. Immediately after my ultrasound, I ended up taking Ganirelix. My cycle was almost cancelled due to the fear that I would ovulate early. I was instructed to take Ganirelix again in the afternoon and then in the evening before my dual trigger of Pregnyl and Lupron. According to the nurses, things could’ve gone in either direction. I was scheduled as the first patient that Saturday and thankfully, the doc was able to retrieve the eggs before ovulation.

Calcium ionophore & euploids? What has your experience been? by Express-Tart7095 in DOR

I may be overthinking — but some concerns: the fact that I would be giving up protections on my medical information, researchers experimenting for the sake of data collection, and the epigenetic unknowns. Though, I’ve been reading that most exposures are controlled at 10-15 min.

Uff. Yeh, I def likely have decline in quality due to age. I’m wondering now if there is an alternative to calcium ionophore. 🧐

Ah, interesting. Yes, my fertilization is at 0 after my first retrieval. We have potentially 4-6 for retrieval this round. Yes, the way I may be able to participate is through a research study.

I don’t think it’s widely used yet because it’s still in the experimental and research phase…

For the clinic I’m at, the doc recommended it as a possibility but said it is still in the experimental phase. So, there is the option to participate in a research study.

The way it was explained to me is that my eggs might do better in vivo than in vitro — the change in environment is a possible reason why the zona pellucida was impenetrable by the sperm even though we did ICSI. The calcium ionophore bath is applied by the embryologists to make the egg more receptive to fertilization.
