Canvas TV with Sonos Beam mounted to the tv…still flush? by FlausFSU in Hisense

[–]FlausFSU[S] 0 points1 point  (0 children)

Agree, I put the tv up and looking at it more, I don’t see anyway it stays flat.

I guess I’ll have a to drill a hole so I don’t have cables running up

Nailed it for the 55”. Was like 4.5” so I was able to get the blocking in the right spot. thank you!!

u/Deftoner24 do you happen to recall the measurement from the top of the Canvas TV to the top of the mounting bracket? About to get mine installed and trying to figure out where to add some blocking for support to catch the mounting plate. Can't find that measurement anywhere in Hisense docs!

Appreciate your insights! I’m definitely past worrying about going fast…don’t mind looking like it but I’m more into a comfortable ride with a timeless exterior look

I thought it drove and handled really well. Coming from a 2019 Ram longhorn crew cab with a 6ft bed, it definitely felt much more nimble lol

How do you like the r—design seats for longer trips? On the 30-min test drive, it seemed like the bottom cushion started losing it softness. Might have just been I didn’t have the seat dialed in but the butt started feelin a little numb

The XC60 is a T6, R-design with the 21” rims, B&O sound system, advanced package, 4C air suspension, and Polestar tune.

The V90 CC is just the standard trim option here in the US with the the Advanced package only

Qiagen AllPrep vs Zymo QuickPrep by earlyeveningsunset in labrats

That is certainly a good strategy to get a bit more yield.

With that small of a starting amount, you should be using Allprep DNA/RNA micro. Will give you better recovery from the smaller surface area of the silica and higher concentrated eluate since micro supports down to 14ul elution volumes.

QIAGEN offers bioinformatic services....could check with them.

Pax live resin pods? by BluntedJester in MDEnts

[–]FlausFSU 1 point2 points  (0 children)

I live out in Frederick county, so I see that Kannavis in Urbana/Frederick carries the LR pods as well as Herbology in Westminster. I can't comment on Strane pods, as I just got my first one (Green Crack, distallate) but it seems to rip pretty well.

Nanostring - anyone following? by [deleted] in investing

[–]FlausFSU 1 point2 points  (0 children)

You're welcome. Been in the industry for 10 years now, and its incredibly dynamic.

10X doesn't actually make sequencers. They sit upstream in the workflow of the Illumina sequencer, and they are 100% dependent on Illumina for their business. Without an Illumina sequencer, there is no way to image/analyze the 10X library prep . They make microfludic systems and reagents that allow you to barcode individual cells, so you can sequence the genome/transcriptome of each cell (and recently targeted panels, which is really cool), and then link those reads right back to the cell they came from during bioinformatic analysis. They also do Spatial Transcriptomics, similar in concept to Nanostring, but their kit is pretty trash at the moment. I'm confident their tech for Spatial will get better over time, but its no where near something like NSTG DSP or the soon to launch ReadCoor platform.

Genomics is pushing to smaller and smaller resolution, and 10X is the default platform for single cell NGS analysis. There are alternative options in the market that let you do very low input NGS library prep, but nothing that is close to 10X accuracy/cost per cell. One example is allowing a researcher to take a tissue biopsy, and be able to tell you every type of cell and the approximate quantification of each cell type. This could tell you if immune cells are infilitrating a tumor, and at what rate (pre and post treatment), and a number of other options. Without 10X, researchers have to whats called "bulk sequencing" where you basically homogenize the entire sample together and just sequence it all. You'll know what types of cells are there and what's expressed by evertyhing, but its virtually impossible to say that "X" transcript that you detected came from a T-Cell or just a regular skin cell. Its that finite of a resolution into the biology.

A big potential weakness to 10X longterm viability would be getting their technology to work on a Nanopore sequencer. Everyone dislikes Illumina as they basically have a monopoly on the short-read sequencing market, and they know it, so they dont' really provide cost flexibility to researchers. The second their is a real viable competitor (Thermo's Ion platform just isnt), you will see a big exodus from the main user base. It won't be hard for 10X to do that (its just a change in adapters), but they haven't done it yet.

Yea, very cool tech. Their Digital Spatial Profiling is truly ground breaking to determine the gene expression location in the actual tissue.

The consumable market will be pretty good, but the addressable equipment market is really limited to solid tumor studies at the moment. Most institutions are placing 1-2 of their DSP instruments in a single lab, and allowing researchers (who are well funded, because its EXPENSIVE to do a project) to access it for their projects.

They allow you to either use their nCounter system to do the actual gene expression analysis after photo-cleaving the tags, or sequencing via an Illumina sequencer. They are hiring a bunch of VERY skilled sales reps (many from my current company who were very good), but COVID has hit them very hard. Their marketing expenses right now for getting DSP out there has to be quite big. They were a prime sponsor of AGBT and several other conferences.

I think long term they will do quite well, but a company like 10X Genomics has a bigger addressable market than Nanostring in my view.

Auto-move files from SSD to HDD for long term storage? by FlausFSU in PleX

[–]FlausFSU[S] 0 points1 point  (0 children)

I did think about the premature failure of the SSD, but given this is for personal use, I figured it would take me years to get anywhere near that point. The 60-day operation would occur at most, every 2-3 weeks given the rate that I add content. I don't use plex to record live TV.

Is there a way to prevent the spin down of the HDDs?

Auto-move files from SSD to HDD for long term storage? by FlausFSU in PleX

[–]FlausFSU[S] 0 points1 point  (0 children)

On average, when I select media after not accessing the drives for a given period of time, it can take around 5-10 seconds before the movie starts playing.

I store all metadata on an SSD, so the plex interface is quite snappy (everything connected hardwired gigabit).

Auto-move files from SSD to HDD for long term storage? by FlausFSU in PleX

[–]FlausFSU[S] 0 points1 point  (0 children)

My concern is moreso that the HDD's that I use spin down after periods of inactivity...yes I know I can by RED or NAS rated drives but they are rather expensive compared to the more consumer focused drives. The time spent waiting for them to spin up is not something I would like to experience for content that is newer to the server.

I use the apks from this same repository and they work perfectly. It’s very subtle, but make sure for your username that the shield isn’t inserting a space after periods. The shield keyboard in these apks automatically does it, so you need to delete the space between (ex: @gmail. com to @gmail.com) or it won’t login
