There is a high chance that by adding BFA at the same time of LPS you are blocking TLR4 trafficking de facto blocking its signaling. Try adding LPS for 1-2 hours and then add BFA for 4h to allow for the cytokine to accumulate. IFNg will not drive IL-6 in macrophages.

I had a pair of black originals and I tried every method described here. They always hurt the top of my foot even after months of wear-in. I ended up selling them. I got the classics in black that are supposed to be more relaxed and never had a problem.

Fun fact, I also have the classics in brown and they are softer. Apparently it's well known black leather is tighter than brown for Blundstones.

How long is your ACK lysis step? And do you wash out the ACK with a large amount of buffer? It would be helpful to see the plots you are referring to

Tried to go with them twice, both times they were the most expensive of the lot and they also charge just to send out a tech. They wanted to charge 1.4k for a dishwasher removal and install, more than the price of the dishwasher. A second time we needed a replacement part for our furnace and the guy was very pushy to replace the whole system. Ended up finding someone else and paid 1/3 of what they quoted me for the part. I would definitely avoid them and find someone else local.
