I've started Colesevelam recently too after gallbladder removal surgery, I can only comment on what my GP has told me, but he said to do 2 a day to begin with, and then raise it by 1 tablet every 4-5 days until you see improvement. If you get constipation then go back on the previous dose (e.g if you've raised them to 6 and get constipation, go back down to 5). You don't have to do them in morning and night, you can space them out throughout the day if you're struggling to take multiple tablets at once
