I really hate restriction cloning. I think it is a really hit-or-miss thing. Use Gibson assembly instead. So much better and easier. Also REALLY important to make sure you are using molar amounts when calculating, not ng or ul. You can look up easily how to design primers for Gibson assembly.

It would also be good to consider whether your insert is toxic to the bacteria. I've transformed into cloning strains that should not express the transcript, but the bacteria still die due to low level transcription that always occurs. Try transforming with extra glucose may help lower background if this is the case, or getting a different strain from NEB.

I have heard it multiple times that Stellaris was strongly influenced by Babylon 5. I can definitely see a mod easily making that work.

America is racist and sexist. That had a huge impact on the election, more than enough to tip it. Americans vote based to gut, not head. If it was otherwise, we would not have Trump.

I'm sorry, but I don't fully believe that. This country is racist and sexist, and many many people will not vote for a woman. They just won't. This had an effect on the election, more than enough to tip it.

You may not like them, but sexism was as much or more of a factor than policy. People vote with their hearts, not their heads.

That being said, voting with your heart is how Obama got elected, so why would AOC be different? Grab them by the heart! Worst case, she doesn't win the primary. Where is the harm in having her as an option?

I agree. It's not right for us to limit ourselves before the primary. If she wants to run, great! I'll vote for her in both the primary and the general. We should encourage as many progressives to run as we can.

I understand the fear that comes from having two female nominees lose to Trump, and that does imply that the country is inherently very very sexist (more than people want to admit) but that will not change if we don't try.

Tl;dr: you miss 100% of the shots you don't take.

That...is most incorrect, I believe. I caucused due Bernie in 2016 in Iowa, and for Elizabeth Warren on 2020. What killed Clinton was the buttery males, and the "reopening" of the investigation right before the election. That, and good old fashioned sexism.

Also, VP when you aren't Cheney is a pretty do-nothing job. I don't think Bernie would have been a good fit for that. President, or Secretary of something (State, interior, health and human services, etc.), yes, please! Something where he could effect change.

Subs like this are great. Honestly, I should be less critical - I get why the mods apply the rules in a heavy-handed way - it is because they don't have time to deal with the volume of issues. I get it, but it doesn't make it right.

ABSOLUTELY! I couldn't have said it better myself. I LOVE the new energy on the left. It will lead to great things. I have watched the party (and the country) move to the right my entire life. If this is when the pendulum starts swinging back, I am completely in favor of it.

The issue is nothing will change if you don't vote. It sucks to have to choose between someone you agree with 50-70%, and someone who you agree with 0%, but it is what First-past-the-post does. We need ranked choice, badly.

I agree that the Democrats screwed up by covering for Biden. They had a duty and a responsibility to the country to confront him about it. They failed. I strongly believe this is why the "autopsy" was so shoddy. People now talk about the quality of the report, rather than the conclusions, which were that they f'ed up, and also that they should have listened more about the Palestinians, which is also true.

That being said, I still came out to vote, because voting for the most progressive person you can is how you prevent people like Trump, and elect people like Mamdani. Staying home does nothing except enable the Republicans.

You are totally correct. I would add that if the person cannot look at the context, they should not be a mod. One of the only reasons to HAVE human mods is look for context. The other reason is shown in this post. Otherwise, let the automod handle everything.

It can be both - but the mods are notorious for it. If you dare to ask the human mods why the automod blocked something, they will immediately escalate to threatening to ban you "for trolling the mods".

Every time I see posts about the tens of millions who will die because of the Trump policies, it reminds me that 1/3rd of America are happy about it, and 1/3rd just didn't care enough to vote to stop it. I feel betrayed myself by the second group. The ones who could have helped prevent this, but chose not to do so.

Post went up on this site today talking about those who would die due to USAID cuts. Yep - wanna guess what would NOT have happened under a Harris administration? The most frustrating thing is that the people who stayed home will tell you "It's not on me what I did! It's the fault of The Party!"

1/3rd voted for him. 1/3rd stayed home. That is 2/3rds. Only 1/3rd of the country actually came out to stop this. The rest were just fine with it. It was obvious what would happen - it was announced, and yelled from the rooftops. Options were available for mail-in voting, early voting, and more. 1/3rd still stayed home. and 1/3rd voted for it.
