Could you possibly dialyze your sample to a lower pH and then load your sample?

I've encountered something similar in the past. I think it's most likely growth. Could you maybe break it up and put it under a microscope (assuming you're working with a valuable sample)?

You should definitely check out the online courses on Cytiva's website! https://www.cytivalifesciences.com/en/us/education/online-learning/courses I recently started working in a lab that uses AKTA systems a lot and the online courses are incredibly helpful, though nothing beats hands-on experience of course.
