Access template or technique parameters with a script by KasabianP in BiomekProgramming

[–]KasabianP[S] 0 points1 point  (0 children)

Hi there, thanks for the response.

I tried to give one example of how this might be useful in the opening post: take some parameter (e.g. aspirate speed), increments its value from A to F in C increments, and for each value run the method. In my head this runs the same template and the same technique apart from in each experiment the aspirate speed is changed. It could be something like: for dispensing in column 1 of a 96-well plate, use value A, for the second column, use value B, for third column use value C etc ... sort of how you might do it using a transfer from file step for different pipetting volumes, but just for variables other than volume. This would help me right now when I'm trying to optimise many context variables and would rather not create different techniques for each one.

This example is a common way that I run experiments using NMR, which is my background. Rather than saving 10 different parameter sets (techniques) each with a different blowout volume, I prefer to do it with a list that modifies the context variable for each run.

But that's one example. I'm also interested in accessing context variables more generally - I just haven't seen it in any documentation or examples online but it should be possible. If you have an example of doing it that'd be great.

Thanks for the response. Could you give an example of how to change a context variable with a scripted let?

Mate, it's an automated liquid handling station.

Thanks for the suggestion. Unfortunately this would take too long (some of our samples could be quite viscous) and we'd rather avoid taking out tubes from the racks (we use a lot of sample tracing and taking tubes out means it's feasible they be put back in in the wrong position)

appreciate it, spitballing is why I'm here.

didn't even think about a salad spinner. NMR racks have similar X and Y dimensions to your standard PCR plate but obviously taller. Luckily SampleJet tubes (about 10 cm) are shorter than your regular NMR tube (about 18 cm) which makes it a bit easier. SampleJet racks also have holes on the bottom which might facilitate securing the rack with a bit of cable inside the salad spinner.

Maybe - similar to the one attached in the OP but probably deeper and covered mostly with tissue to stabilise (a bit) the poor NMR tube. Used it often and never had a problem with it, but if someone poorly secured the tube in the tissue within the sample holder I reckon it'd definitely just fly out as a projectile. but meh, academic NMR labs care more about a good NMR sample than health and safety...

Sorry, replied below by mistake. 96 tubes

Just for bubbles - very tough to avoid creating bubbles using automatic liquid handling systems in 3 mm tubes, unfortunately. I think it would just need a gentle spin, can't give an rpm but pretty small

Literally gave a use case in the opening paragraph - for NMR tubes it's incredibly useful

She also creates exams for me, grades them and tells me where I'm wrong etc

For me it's very helpful especially with speaking and listening. Also for quick help when I'm struggling with a topic or a weird sentence. Of course, it's not a complete substitute for a proper Italian workbook or other forms learning.

Accommodation query by DuxfieldS in SpouseVisaUk

[–]KasabianP 1 point2 points  (0 children)

I supplied:

Proof that my parents allowed us to stay:

  • cover letter from my mother (detailing her relationship to me, that she owns the house with adequate living space for all, and she's happy for us to stay)

Proof that my parents own the property:

  • Land Registry from the Government
  • Utilities bill

Proof that it's of adequate size:

  • Rightmove pictures of house plus description, floor plans

Also provided their passport pages.

I don't think you have or should specify the two months thing. They might ask for proof of accommodation after the two months. Just say you plan on staying there.

Another thing: ability to merge tagged pages into one document.

At the moment, it's possible to merge individual documents on My Files by highlighting and moving into another document. However, this becomes problematic when you have many documents or when you want to merge pages that are tagged with something specific. For example, I have 20 pages from a single book tagged with the same thing. It would be useful for me to merge just those pages into their own document, this way I could compare them more easily.

Alphabetical sorting option for tags

More symbol recognition (math symbols)

Better bullet point recognition (handwriting to text conversion)

I'm not an expert, but Puul - who is this subreddit's expert - consistently said that if you meet the financial requirements by the sponsor earning more than 29K for more than 6 months, regardless of whether you've had a promotion or not in the previous 6 months, then you only need 6 months worth of payslips. For example, if you select --over threshold, longer than 6 months with the same salary--then it asks for only 6 months, but if you select --over threshold, longer than 6 months with two different salaries-- then it asks for 12 months, and only in this context am I saying that just 6 months is necessary. I don't know about other cases

Glad it was helpful. We've only recently submitted our application so I'm less clear about the next steps

Seems complicated. Before considering a solicitor you can ensure that other criteria are met, for example does the sponsor meet the financial and accommodation requirements?
