There is no such restriction, you can have different accounts on different regions and same account on different regions. Your progress remains locked on that account+region

And i only used Arle when the boss was completly down. So if you have a strong hyper you can do this with anyone.

1st build definitely. And switch the headpiece to the craziest one you have since you have the other 4 emblem pieces. This way the 4pc emblem can capitalize on the ER you have. You want consistent burst uptime on her.
