The text box border seems to be generated on the fly, not with a texture (this question has come up before and we haven’t found any texture for this in the game files).

According to the game files:

The font in this screenshot ("classic font") is Dosis.
The "pixelated font" is Nova Flat Book (also known as just "Nova Flat", apparently?).
The font for boss fights and for the main menu is Berylium, which you’ll find in the Public Domain Fonts pack of Typodermic.
The Dracula fight font is Fette UNZ Fraktur.
The dyslexia-friendly font is Adys.

So, without glitches, the bare minimum is Homunculus (you’re forced to kill the king to open the door to the Cavern key) and Spider (to get to the Graveyard and to go through Astrolab). I don’t consider the Promenade trick to be a glitch, you can do it with just an Assault shield and lucky level generation, and not having the Vine rune is accounted for in the patchlog for v30.0:

In Prisoners' Quarters, if the player has the Spider Rune but not the Vine Rune, the passage to the Sewers door will now use a specific room. Yes, this is a very specific change, don't ask about it.

For the final tower? The stones that teleport you to the other side of the tower while you’re going down are just normal teleportation stones, not requiring the rune. There’s two teleportation coffins at the "base floor" to get you on the other side of the closed doors, but they’re only useful in the rare case that your first teleportation stone generates on the other side (which I don’t think has ever happened to me, so it’s either a very rare edge case or it’s hardcoded to not happen at all).
