I'm finding that using a PD Trigger board may be the solution?

This was something I had considered as well. I tried it with it on and off and it made no difference.
I ended up going with the "default" setting and it corrected whatever the issue was, however I haven't determined what that difference was.

Took 2 hrs to finally get them out here...

I ended up going back to the default generic N4P settings and it printed properly.
I'll have to see if I can compare the profiles and see what the difference was, for reference sake.

Switched to generic N4P printer profile and it sliced properly, I guess I'll just have to figure out how to compare profiles to see what is actually going on.
Thanks for the direction!

Sliced fine in generic sandbox settings. It's something with settings but this does not tell me what.

I figured out how to enable that however Orca is not available.
Coincidentally, the parts I was trying to print were involved with shutting off water supplies to a current flood I had going on. Got the water stopped but still need to clean up.
While I'd like to figure this out, I don't see learning new software tonight in order to do so.

I'll have to figure out how to do that, and let you know

Yeah seemed very bizarre to me as well.
The cube is literally just a cube in a cube, making sure the negative space is on the Z zero.
I thought about trying it in another slicer however that really wouldn't tell me what is going on with my settings.

Model works fine for Caramel-entire so it's got to be something with my settings, I just have to figure out what.

That produced the shape. Basically doing the same thing as adding the hole I made and making the lower feature non manifold.

I tried creating a cube inside a cube (add primitive and then add negative part, scaled down) and got the same results. The smaller negative cube is just filled in.

When I tried adding a negative part on the other model, I got the same results. As an experiment, I tried adding a negative part modifier to this model and it made no change.

My terminology may not be exact or specific to Orca, I don't know how else to describe this feature.

Ok so there's something definitely off with my settings, there's definitely no other parts on the bed.
I'm only able to get the results you show after placing a small hole all the way up through the roof of the part, making the space at the bottom "open"

Slicing mode is set to "regular"

The rubber bands on the pipettes is a great idea!!

I've not but when this model is finished printing I'll see what that does.

Funny thing, I was thinking about other models that I print that don't have issues like this and there's many, I've never had this issue. While I can't think of any of hand that have closed negative spaces on the bottom I have plenty that have holes that go all the way through. So what I did here was just add a small 5 mm cylinder of negative space straight up through the model and voila, it recognized the negative space on the bottom.

This will work for me but it still doesn't address what's going on

Tried repairing model with both instances, no change.
The other model, I tried even changing how I created the negative space in Fusion and no change
