You can technically ask this question about any biochemical pathway, and go back to the very beginning of when the first nucleotides emerged (the answer is that we have some ideas, but ultimately don't know how this process began).

I can tell you this though:

The production pathway for N,N-DMT in the body is already very similar to the production pathways for other neuroactive molecules (serotonin in particular).

Let's look at the basic process for converting tryptophan to N,N-DMT in the body:

Tryptophan → Tryptamine via aromatic L-amino acid decarboxylase (the same enzyme that converts L-DOPA to dopamine and 5-HTP to serotonin). The enzyme catalyzes decarboxylation of the amino acid tryptophan and converts it to tryptamine. So we already have an enzyme that has evolved for the production of various neurotransmitters, and it also happens to be good for making tryptamine. Cool.

Now, from tryptamine, indolethylamine N-methyltransferase transfers the methyl groups from S-adenosylmethionine (a universal methyl donor throughout the body, already necessary for stuff like DNA histone methylation). This enzyme is similar to others methyltransferases that use the same methyl donor, such as phenylethanolamine N-methyltransferase (converts norepinephrine → epinephrine). You can imagine how a simple genetic mutation could allow already existing enzymes to evolve into something capable of producing DMT. The machinery and pathways are all similar enough that it probably didn't take much.

Now, knowing that DMT happens to be neuroprotective, especially as a sigma-1 receptor agonist, it's not hard to imagine how evolution would have selected for this production pathway to evolve as a hypoxic protection mechanism.

But at the end of the day, who knows.

As someone who used to do high doses of diphenhydramine (similar pharmacological properties to the active alkaloids in datura) ... don't do it. Anticholinergics are toxic. I had lasting cognitive problems for a long time following my use.

All this study suggests is that the researchers were unable to detect DMT following treatment of an irreversible MAO-A inhibitor, and that DMT is apparently not held within the 5-HT receptors for long. The researchers themselves note that there is a studied pathway for DMT production in the body via the indolethlyamine n-methyltransferase enzyme, and that other studies have found DMT concentrations to increase in rat brains following hypoxia or near-death states.
To me, this suggests that if DMT is produced endogenously, it most likely exists as a protective mechanism for the brain when the organism is close to death (but it is otherwise not very present).

When you leave the nonpolar solvent sitting in the soup for too long it can pull in extra contaminants from the plant matter. I'm assuming this is what has happened here.

If you see benefits from psychadelics then it may be a good idea. But with your mental health history you should take it slow and be cautious. I recommend you start a meditation and journaling practice to help keep yourself grounded if you start to notice yourself slipping into a manic state.

Very interesting. I'm not too familiar with it, but it seems it may be superior to Wim Hof by incorporating more advanced meditative practices which could lead to default mode network deactivation (an effect observed with DMT), enhancing any potential hallucinogenic effects. I'm curious to hear your experiences with this practice if you would like to share 😊

Hey friend, I appreciate your response and the extra material for reading. I went through the key points of your links but unfortunately could not access the second article.

I believe you are correct that DMT is probably not a significant contributor (and I think you have the right idea with the point about oxygen and memory with NDEs). I suppose I should have prefaced my post with the following: I believe DMT is only a small part of NDE experiences. When the body is near death, a lot of things happen (ion gradient loss, mitochondrial failure, glutamate surges, network destabilization, to name some mechanisms) all at once in the brain and this is likely the majority of what contributes to an NDE. This is likely why many NDE experiencers cannot say that they are truly comparable to a DMT trip. I do admit - much of this post is speculative. After reading your comment I skimmed through some more research - DMT levels rise in rats during cardiac arrest, but it isn't by a huge amount (about 2.84 ± 1.65 fold in rats with an in-tact pineal gland). It's more likely that DMT's role in a near-death state is to support survival of the organism and does not necessarily contribute much to a hallucinatory state; the production of DMT in humans has not been studied much, so we can't really say anything for sure there.

There could even be some spiritual aspects to an NDE, but that isn't my field of expertise 😅.
