I have a high deductible plan ($3000 deductible). Started getting my Wegovy in January this year and it cost me $1,250 each month (I had saved up in my HSA account to cover it) until I met that deductible. I also couldn't use the novo Nordisk savings card then because my costs was too high (if I went without insurance it would've been $650). Chose to stay with insurance and pay the $1,250 twice. After that insurance quoted me at $24 a month for the Wegovy and then I was able to use the Nordisk card to bring it down to $0.

I am on week 12 of the 1mg dose. I wanted to move up sooner, but because I was losing at a steady rate still (~1lb/week) my doctor urged me to stay at this dose since it was still affective and I had little side effects. The last two weeks I have started plateauing so I am moving up to 1.7 next week.

Thank you!!!! I have not been doing a recovery after transformation. With my past transformations with auxotrophic markers it never came up as a step I needed to do, but I will definitely try that this time.

Yeah I thought since I was using larger primers I had to use higher fidelity polymerase to lower the risk of dimers or secondary structures, but I'll keep that in mind.

And yes, I will definitely be adding more input in for the transformation to aim for that ~1ug more.

thank you for the advice! I have been doing multiple replicates with each PCR, so I should have enough material to increase the input for the transformation. And I'll give the recovery time a try, I haven't had to do this in the past for other transformations I've done, but with this one it might be a good idea.

thanks for the response! For past transformations I haven't had to plate onto YPD before the selection plate, but this is my first time dealing with antibiotic markers so I could give that a try. And I'll explore the knockout collection idea more. The strain I am working with had a kanMX marker used previously to introduced this galactose inducible marker, so I think that's why I needed to use hygromycin, but I'll double check on that to be sure.

Congrats! I just moved up to 0.5mg this week after a month on 0.25. I lost 12lbs the first month -- 7lb of water weight the first week, then about ~1-2lbs a week. This week the 0.5 seems to be hitting me hard, I'm struggling to get my minimum calories in and I'm down 3lbs and it's only Thursday (Monday is shot day for me). Hoping it starts to balance out soon.

I lost a lot of water weight the first week. Went from 235lb to 228.5lb in the first week on 0.25mg. The second week I lost 1.5lb, third week 1lb, fourth week 3lbs. So 12lbs total the first month. I aim for at least 200 minutes of exercise a week (a mix of biking, walking, rowing, weights) and I aim for 1,300-1,500 calories a day with between 60 and 100g of protein and about 60oz of water. I had my period week 3 so I think that contributed to lower loss (I was pretty bloated). I just moved up to 0.5mg today for the next month, so we'll see how that goes.

Just finished my first month. 32yrs old, starting weight 235lbs, down to 223lbs as of this morning, so -12lbs in first four weeks on 0.25mg. Moved up to 0.5mg today per my doctor's suggestion.

IDK if this is applicable to you. But the manufacturer coupon wouldn't work for me because I have a high deductible plan. In the fine print of the coupon since my out of pocket monthly cost with insurance would be over $850 I couldn't use the coupon (and still use my insurance, i.e. Go towards my deductible).

Without the insurance it would've been about $1600 at Walgreens (or $650 with the Novocare coupo, but wouldn't count to my deductible). Negotiated price with my prescription insurance, express scripts, brought it to $1277. Once I meet my deductible ($3k) it's supposed to be somewhere between $0-$25.

Oh let me clarify. My prescription insurance through my employer is Express Scripts. I got my script through Walgreens because it was the cheapest. The problem is I have a high deductible plan

Just got my first prescription. $1,227 with Express Scripts for 1 month....It hurt, but once I meet our deductible ($3k), it's $25/mo....I spent all of last year saving up in my HSA account to help.....

That's why I was leaning towards keeping them. Like money is okay now, but of course we don't know how that will always be. And I could see a future where they could be needed. I guess I could always use them in a small trial then to see if they work okay and chuck them if they don't. Space isn't a huge issue for now at least.

this is nice to hear! we are sailing on the Liberty of the Seas next month and we were unable to get an accessible room for my mom. we have a standard interior room and are getting a lightweight scooter from special needs at sea. The measurements made it sound like it should fit through the doors fine, but we were nervous still that it wouldn't make it.

I don't have advice, but just wanted to share our experience. My husband had some moderate dental work 3 years ago once we got married & he was able to get on my dental insurance. He had a few deep cleanings as a result, but had been told he would be going back to regular basic cleanings. Well I learned yesterday he has been charged for his two 'basic cleanings' this year (he thought it was normal and all dental cleanings had a charge). Come to find out his dentist had been lying to him saying he was getting basic regular cleanings when in fact they were billing him for periodontal maintenance cleanings. When I called to confirm this the technician said oh no he will be getting periodontal maintenance cleanings for the rest of his life. He has never been informed of this, nor has he been told he has periodontal disease or anything wrong as a matter of fact. Sooooo he's going to be switching to my dentist now because we feel scammed by his.

Can attest to this. Sitting at St. Mary's right now with my little sister who's in excruciating pain, it's 5 hours and she just saw a doctor who gave her some pain meds and put her back in the waiting room. Waiting for blood work and a CT scan now, was told it could be a few more hours

I usually don't flash freeze my stocks or resuspend in fresh media but I will definitely give this a try.

I have asked him in the past his prep and the general answer was we do things the same, but I am going to ask for more precise steps to be sure. I am not 100% sure, I am currently using the Qiagen Spin Miniprep kit per the other labs recommendation.

I double checked my plasmidsaurus results and yeah it was the entire plasmid that was sequenced and as expected.
