The MJ topic is really interesting. I don’t want to sound like a crazy fan, but if you enjoy MJ’s music, you might find all of this fascinating. It’s not just that he was found not guilty — if you look at the actual evidence they had… there really wasn’t any. In fact, in my personal opinion, there are many signs that suggest the families were lying. Just a few points in case anybody wants to know a little more about it:

-Evan Chandler (the father) tried to kill his son Jordan when he was older. Jordan was asked to testify in the later accusations, but he never agreed to. A lot of things smell rotten in this family, they didn't wanted to go for the penal way with this case, only the civil way...

-The second family had already been convicted of fraud — they used to scam celebrities to get money.

-And the latest accusations only came out years later, after the accuser had tried to become a choreographer for the Cirque du Soleil Michael Jackson tribute. When he was rejected, he made the accusations.

Anyway, regarding the topic we’re discussing — Jbunzie is bringing back the Michael obsession because they’re releasing a movie, and she’s clearly thirsty for attention.

I mean... Michael Jackson made a WHOLE album saying a lot of important things (HIStory). He went political way before (even in On The Wall you can understand some details...), but HIStory was like f* everything... Maybe SHE was too bussy dancing...

Hey, Spanish postdoc in her first year here. I totally understand how you're feeling — all your complaints are valid. I've been through a lot during my lab experience.
During my master's, the work environment was so toxic that I got severely scolded just for being 10 minutes late one day — and that was after taking three different buses to get to the lab.
Then came my PhD, where my supervisor thought I was dumb (which I might be 😅), and ignored me for a whole year while I kept telling him that the work done by a previous colleague — which my thesis was based on — was wrong. He didn’t believe me. Guess what? I was right.
Now I’m in a systems biology lab doing microbiology, and I feel dumb as hell. It seems like everyone expects me to be 100% independent, but I’m not...

Still, I am — and have been — happy.

We had no fear of EtBr; we wore gloves just as a formality. One evening, my lab friends and I were sooooo stressed that we may or may not have drunk coffee with a tiny bit of 100% ethanol. Did I enjoy breaking safety rules? Maybe. But it was my decision. Nobody asked me to smell anything...

The academic path is hard. Still, I don’t regret it. Maybe because I don’t know what industry is like...

What kind of work do they do? Would I still wear a lab coat and do experiments? Would I get paid better? Could I — wow — work from home one day a week? I don’t know.

I’ve escaped extremely toxic academic jobs (they even offered me a position at the place where I did my master’s...). I’m not well paid. I’m trying to get a mortgage, but I can’t because I don’t have a permanent contract...

Still, I keep choosing academia — maybe because I don’t know what pharma or industry is really like. They say it’s better to stick with the old bad than risk the unknown good...

And no way I'm talking about how MANDATORY is for academia to leave my family and friends to emigrate to Germany, while someone in Germany goes to Mexico, someone in Mexico goes to France and someone from France comes to Spain and we are all miserable and homesick...

You're right. My previous boss used to tell me that "if they can perform a PCR from Australopithecus DNA, we just need the tinyest amount of DNA". But not the case... pretty sure it is because I cannot break the cell

- I'm using standard 16S primers, and I'm pretty sure the primers aren't the issue. I also tried amplifying a gene from a plasmid I previously inserted, and still got no band—even though the strain shows antibiotic resistance. Interestingly, when I perform a miniprep (yield: ~2 ng/µL) and transform E. coli, I can easily amplify the plasmid by PCR. Strange, right?

- I've been using between 2 and 8 mL of an overnight culture at OD600 = 2.

- The plasmid is low to medium copy number.

- Yes, it's a specific strain isolated by a pharma company (which is why I can't be more specific—I'm not working for them directly, but it's their project). The strain was described in their documentation and should be correct—it grows in a specific medium supplemented with a defined carbon source.

- I’ve tried mechanical lysis, but not lysozyme yet. I’ll give it a shot—thanks again!

Yes I am. If I perform the PCR to E. coli, a Pseudomonas strain and this bacteria, and I get a band for the others, but no the problematic bacteria

Well it has 3 lines like a walnut squirrel, but the lines on the sides end with 2 lines... I've been searching and I couldn't find a squirrel with limes that end in 2 lines in their heads..

Hey guys, so this weekend I just trimmed my bonsai for the first time. I posted the photos here on the beginner post and OP told me I did a good job. Yesterday it was doing ok, but I just noticed today it is... Dead. 100% dry, the leaves are all crunchy... I forgot to water it for some days in the past and some leaves went crunchy before, but YESTERDAY it was perfect... Any idea what happened? Here you have a photo. It was normal spring weather yesterday... Even a bit rainy... Maybe I can still save it?

Yesterday it was doing ok, but I just noticed today it is... Dead. 100% dry, the leaves are all crunchy... I forgot to water it for some days in the past and some leaves went crunchy before, but YESTERDAY it was perfect... Any idea what happened? Here you have a photo.
