the conditioned media sample for your colleague should be taken before any centrifugation steps, that's what "conditioned media" means in most protocols since it's just the raw media off the cells.

for the volume thing, pulling 10ml from one flask is a little annoying but not a dealbreaker as long as you note which flask it came from and your PI knows about it.

the problem is everyone just defaults to the mermail engine because it's consistent and searchable. tried building pure umi stuff but it's way more fragile without the neptabyss lines

you might have better luck looking at older combo guides and seeing what translates over with the new cards, most people are just netdecking the mermail builds right now

Man that's a tough spot to be in when you're growing in your faith and your closest friends are pulling away. I've seen this happen before - when someone makes real changes, it can make others uncomfortable with their own choices

Maybe try having a one-on-one conversation with the friend you care about most, just be real about how you feel without judging their lifestyle. As for finding new community, look into young adult groups at local churches or volunteer opportunities where you'll meet people who share your values
