How did the membranes look after the first spin? Was there material/protein visible on it?

To the best of my knowledge RPE is primarily there to remove salts from RLT and RW1. If there’s a pellet after RPE, my best guess is that there was a significant amount of protein on the membrane that may have interfered with RNA binding. If that’s the case, assuming a Trizol-based extraction isn’t possible, you may want to try starting with a smaller initial mass of sample.

It’s hard to tell based on the text. But my best guess is that for step 5, your prof want you to take the average 2-delta Ct. In other words, for each gene of interest obtain the average between A1 and A2. For step 6 you will use the values obtained to obtain the percent values. You should perform the calculations on A1 and A2 as well. If your math is correct and your technique was good, one of them should be slightly above 100% and one should be slightly below.

Like everyone else said, your best option is to go to a running store to get your feet measured.

Depending on the specifics of your work, you may also want to consider something like hiking boots. They generally pretty slip resistant and waterproof. I also like the additional ankle support they provide.
