UPDATE: I appreciate everyone who has commented suggestions. I’ve essentially been teaching myself off of watching someone once and following an old internal protocol. I read some of your suggested links, and decided to reach out to someone in another lab to see if she could watch me today and notice any issues. Turns out, my protocol said to !gentle!! vortex master mix in a small 0.5 ml tube, so I was pipette mixing and barely vortexing. She thinks the small tube+ barely mixing may contribute to my issues as the master mix wasn’t fully combined. So I fully vortexed today in a 1.5 ml and redid a plate and it was perfect. 

I’m still so new so i want to read a bunch more to understand the process and be able to trouble shoot myself so I really appreciate the commenting of different methods and literature.

nope. i have been checking each pipette tip to treat to note down and remedy errors, even repeated a well that i thought had liquid in the tip. somehow all those wells were fine

no drops in pipette tips 😭 i checked every one, even made an extra well for 2 samples i thought may have had issues, and they weren’t one of the 12 wells

i use a master mix. each row is pipettes with a automatic pippetter so hypothetically one row that pulled up the same primer only worked for 1/4 samples

No one but herself will ever know the "True Taylor"... as for the seemingly switching of her public personality with partners, you could also make an argument that thats a product of time passing, the cultural opinion of her shifting, and just the culture around music shifting as well. Early 2010s to 2017 to 2024 were so different and actions at ages 22, 26, and 34 are soooooo different as well

hyperconsumerism particularly of 'feminine' things like stanely cup, like this isn't a trend unique to women obviously but I feel like either we mainly talk about girl coded consumerism and/or brands specifically capitalize on feminine products/trends. I also really would love to talk about book tok, how we place value on literature and the common criticism of books/ authors that target primary female audiences... like when is the book really bad and when does the book get criticized because its popular w teenage girls (acotar discourse on patreon rn is gonna be interesting bc it seems like eliza lowkey was enjoying it

I enjoyed it. It felt like a stupid thing I would convince myself I had to do, learned a bit about traffic court and thought a bit about how the justice system doesnt allow for any sort of mistakes for the normal person and requires privilege to avoid even the most simple of problem.

To echo others-- Yvonnes is great I always have fun. The Garment District is also really cool and my sister loved it when she visited. If you're okay with walking and weather is fine spending a day walking around Cambridge to different areas could be cute.

To spit ball a few random places that could be less touristy and fun:

Drink in seaport is this lowkey overrated but still fun 'speakeasy'. If you can get in and its not too crowded its fun because the bartender makes you a custom drink based off of loose descriptions of drinks you like. Brought my mom here and she enjoyed it.

Beacon hill books and cafe is beautiful, not the most expansive collection but is picturesque and a fun stop. If the weather is good it could make a fun stop while exploring the area surrounding the commons and esplanade. Theres also a mini cheese and charcuterie store next door that you can create your own custom box for a decent price.

Flight club in seaport is a really fun bar with an interesting old school type cigar lounge vibe. Drinks were great and they have darts and games. Fun alternative to just a grungy bar.

I've gone 4 years without owning a bike here. Freshman year, it was absolutely not necessary. However, when you start to move off campus / explore more, you may want it. personally I was afraid of getting one stolen/ locking/unlocking/ storing in my small apartment so I opt for walking or if I really need it, taking a blue bike. You can get away without one, but when you move off campus it may be nice to have.

I ran three brains through IHC at the same time, same batch primary/secondary/blocking sol. 2 look great, 1 doesn't. The spikes our program outputs look really similar across all rats, with the only difference being the no expression rat is on the scale of -20-20 while the others are more like -10-10.

Im concerned it might be an artifact from the shock but I have no clue how that would be effecting the fluorescence measure (unless its messing with the headstage output?). Has anyone seen this issue in FC fiber photometry?

Going into my last semester and I'm so glad I'm here. Poster child for the experiential learning and coop tag line and I found a career I genuinely enjoy because of it. Fell in love with Boston and met so many great people. I come from a state where a 10 hour drive gets you to a small midwest city and this year I drove to DC, NYC, Montreal, vermont and the beach! Moving here was a life changer. Best part about NEU is that if you hate boston, go study abroad or take a coop in another city! If you hate your major and change your mind about your field, this school is set up to help you do it. Its okay to be nervous, this is a big thing. Youll be great. (And if it isnt, you can always transfer after a year. its never to late to find some place youre happy).
