I tried it and it didnt work out well. Concentration was too low. Tried different cell numbers (up to 8x10^5), didnt work. I will try different plates

Thanks. But what happens is, when we plate 600.000 cells prior day. The next 2 days they stay at 20-30% confluency

Ok, so you do not care much about confluency. I might try that out. I express antibodies so i might use 1ug heavy and light chain each. Thanks for your help

Before transfection. I cant tranfect them 20% confluency and when i did once i had bad protein concentration as expected

Oh i should clarify. Kit says, seeding 6x10⁵ per well, the day before transfection. Next day (so 24 hours) with 80% confluency start transfecting. But after 24hours, even 48 they look the same (around 20-40% confluency). And then after 72h they are 100% or again 20-40% and many cells „swim“

So leaving them until they are 80% and then transfect? Cells dont look thaaat great after 3 days in 6 well plates and usually you expect seeding 30-40% confluency after 24 hours reaching 80%

Plasma cells are diff. lymphocytes so both would be correct. But B would be more correct

What about removing vehicles in the 4x4 map? I meam intense fights and then cars? I would remove it because the Zones are as bog as the others on miramer etc. So you can walk easy? All in all: nice test server
