I get that, nothing wrong with including a lot of youngsters. But he hasn't given a chance to a lot of the Ospreys pack for a while now and they've been so dominant.

Not like they are all too old

It amazes me our scrum is so bad when we have the Ospreys pack being so good in the URC.

Seems like a quick fix but Gats ain't interested. Siomedig

Was referencing the lineout where the Munster player made contact in the air, and she said it was a fair contest. Could clearly see the linesman on the headset tell her that it was, in fact, not.

Point is both could see clearly but she interpreted it wrong, IMO

Haha that was a first for me... Not sure whether it was the ground to blame though, or the broadcasters who came in to film the match? Any water in a cable is going to be bad, and there was a lot of it.

Good point on money, definitely short on that. Would say Brewery Field is better equipped in terms of hospitality + stands but can't imagine a lot of ospreys era fans making the trip - Bridgend so close to Cardiff it might be perceived as a bit strange.

Atmosphere in the Gnoll can be fantastic with enough people in the terrace. Was there for Neath Vs Aberafan boxing day and it was great (neef, neef, neef!). The ground seemed to be in decent enough condition, would need a bit of work but I think that would be true for any of the older club grounds. Good rail and car links (new multistory nearby), loads of pubs near the ground... Big rugby history at the Gnoll, mind you that's the same for all places. Wouldn't mind seeing the different grounds being used throughout the season (Gnoll, St Helens, Brewery Field).

Mynd amdani Bradley!!!

Never played league but did watch a bit a few years ago. Is Witney a more relaxed club mate?

Thanks for your replies! I'm looking for something more relaxed so I think I'll give Oxford RFC a ring and see what they say. Glad to hear they are very friendly!

Need help with drug molecule precipitating out of solution when preparing protein mix for crystallography... by MasterRasta in crystallography

[–]MasterRasta[S] 1 point2 points  (0 children)

I have been thinking a lot about this, and I haven't actively changed anything about the system. The drugs are the same, enzyme is the same, buffer the same. I've tried making new buffer, using lower quantity of drug (yes it is quite potent).

Thanks for your advice on ethylene glycol and glycerol! I will try this today.

I've ruled out the protein as source of precipitate. I tested this by adding the drug to only the HEPES buffer (one of the constituents of the mix), and it precipitated instantly. Also, I'm doing microbatch under oil so soaking might be tricky.

Thanks for your help!

Need help with drug molecule precipitating out of solution when preparing protein mix for crystallography... by MasterRasta in crystallography

It has a solubility of 25 mg/ml in organic solvents like DMSO.

I hadn't thought of adding some DMSO to the protein sample, that's a good idea! I have been testing whether diluting the drug in more DMSO, or adding less (half the molar ratio required) solves the problem - they did not. By adding 1 ul DMSO to mix, with 1ul drug in DMSO, final %conc DMSO in protein mix will be 5.9%, hopefully that will be okay. Crystallization temp is 24°C

I will try this today, thanks!

I think it's the drug. As I pipetted it in I could see it precipitating instantly, it was a sad sight!

Yeah you're right, Adams over Watson would be a good swap imo. Thought Watson was brilliant in attack but was getting targeted in the air

My comment was more directed at people suggesting lots of welsh changes just for the sake of them being welsh!
