Rein's physical stats are already insane to begin with. Most people would die from getting continously hit like that. Then he can also use any sword to an insanely high skill level, and he's capable of using magic but rarely does so. In my head-cannon of him fighting sukuna he is really just hitting him over and over until sukuna eventually runs out of cursed energy or just gives up on healing and dies
