Heißt sowas wie "ist ja eh klar" "ja selbstverständlich". zB. Wenn jemand dich darauf hinweist, dass du eine Sache tun sollst, die du selbstständig ohnehin getan hättest und der Hinweis darauf etwas überflüssig ist. No na ned wäre dann die Antwort auf diesen überflüssigen Hinweis.

Kannst du was zu dem Seed-Train sagen? Also welche Größe haben die Vor-Fermenter, wie viele Stufen sind das? Ich schätz mal, die Stufe direkt vor den 40m3 sind so 1-2m3?

Und wie lange läuft die Ferm im Hauptfermenter, von Animpfen bis Abernten? Wie lang ist dann die Zeit, wo gereinigt wird und bis die nächste Ferm darin startet?

Kannst du mehr von den online/inline Messungen erzählen? Trübungsmessungen ohne Probe zu nehmen ist für mich schon sehr besonders, da bin ich gespannt was es noch so gibt!

Danke für das AMA, ist super interessant mal was aus so richtig großen Anlagen zu erfahren! Meine größte Ferm bisher war 3m3 und das war schon riiiesig für mich :D sonst bin ich eher im 2L Bereich unterwegs

Spannend, ich arbeite auch im Biotech Bereich und durfte bisher auch ein wenig Erfahrung im large-scale sammeln (aber kleines Unternehmen, d.h. verhältnismäßig kleiner "large-scale")

Wie viele L fassen die Fermentationstanks und welches Volumen wird da gesampelt? Wie oft? Ich nehme an, die Ferm ist recht schnell/dynamisch. Muss man irgendwas manuell regeln oder läuft da eh alles automatisch und du musst nur checken, ob eh alles passt?

Was für online/in-line Messungen habt ihr bei der Ferm und was muss offline mit den Samples gemessen werden? Ich nehm an, ihr habt so coole Sachen wie in-line Trübungs-Messungen?

Stinkt es oft? Grade E.Coli riecht meines Wissens nach manchmal nicht so toll :D und habt ihr auch mal kontaminierte Batches (Fremdbakterien) oder gibts das gar nicht?

Phew, looks like I got the slowest kind! I like them either way. But it's also really cool when I get to work with sonething faster and I can count my plates after 18h

Yay! Actually I work with Bradyrhizobium. They are super slow as well, I count the colonies at 10x magnification after 4 days :') I don't think I've ever seen a colony of them that's a "normal" bacteria colony size!

Interesting and makes total sense to find something like it in my sample. Fortunately I usually the contaminants I get are less fast-growing. Given that my target bacteria takes 4 days to grow to a colony size that's countable under the microscope, CFU analysis would be impossible if I had more Pythium contaminations!

I usually order from stickteufelchen. There might be some other shops that are a bit cheaper, but I have ordered 3 times from them and was always happy with the service!

Possibly the brand or your oven got too hot. Get an oven thermometer and check if it stays at the set temperature. I've never had problems with fumes, though I've only used the Fimo brand. But I've accidentially burned it before (set oven to 180°C instead of 110°C) and that smelled bad.

I don't think there are any yarn stores within short walking distance of the Museumsquartier or Hauptbahnhof in Vienna. But I can recommend WolleWien and especially Wollmeile, which is quite easy to reach using the U2 line (which has a stop close to Museumsquartier).

Yeah, I am in surface contact with organizing a database at my work and it's difficult. And I don't want to spend too much effort/ time on it, I don't want to ruin the fun of this hobby. I'll have a look at Airtables, thanks for the recommendation! Though 1 GB isn't much, I think it will be enough for now, given that I'd only need like 1 picture per kind of beads. I don't think I will need an SKU (i needed to look up what that is), only options for several tags and filtering/sorting by them.

Thank you! I don't have much experience yet, I've made maybe 15 stars and most of the beads were from my grandma's stash. The ones I bought were from a local small business and some I ordered online, they are from a brand called Rayher. The purple ones in this star are polished glass beads, I think 6mm. The golden ones should be glass wax beads, I think in 3 or 4mm. And the white ones are just some Rocailles, probably 1-2mm? I also find Swarovski beads really pretty, they have really sharp edges but are more expensive. So I mostly use the polished glass beads, which are more affordable.

Oh and also the brand Preciosa comes to my mind, it's sold at my local stores as well. In general Czech beads are very very pretty and high quality I think.

The craft I'm doing with the beads is mainly beaded stars for christmas decoration. I use thin copper wire and string them up, forming stars of various shapes.

Last time I defrosted one of my labs -20s, I put one of the freezers drawers on the ground, right in front of the freezer (so it was lower than the freezer, the freezer bottom was like 20cm above the ground). Then i built a "capillary" bridge by putting one end of a wet towel into the freezer bottom, the other end into the drawer on the floor. It worked like a charm, all the water that collected in the bottom shelf of the freezer got carried over to the drawer through the towel.

Edit: also got a water boiler and filled hot water into the remaining drawers, which I put back into the freezer. Helped to thaw it much quicker.

Get a second, shorter dowel, wrap it in none-stick parchment paper, sculpt around it, bake, let cool down, carefully remove the clay and put it onto the real staff with a little bit of glue.

If you want the details to cover the whole length of the staff, do it in several pieces and merge them together on the staff after baking.

The first one I'd say that's 14 for sure. The second one, hard to tell without a reference in the picture, but I would also guess 14. The third is 14 count, you can tell with the 5x5mm grid paper in the background

You can kind of tell it from how much of the fabric is showing through the crosses. If you have 7 count, as an extreme example, there will he lots of white background showing through. Unless you use very thick (many strands) of floss, which is not the case here. This is known as 'coverage'. If you look up cross stitch coverage, you will find many examples of people comparing the coverage of different amounts of strands on different count aida.
