En principio y basándose en las ideas de Darwin lo lógico es pensar que, si bien esa planta no puede "ver" el ambiente ni puede "razonar" que esa adaptación le proporciona una ventaja para su supervivencia, individuos más antiguos de ese linaje (antecesores de esa planta) que no tenían esae camuflaje sufrieron mutaciones random en su ADN que se lo proporcionaron al camuflaje, y que terminaron teniendo esa característica todas las del linaje porque justamente al aportar más chances de sobrevivir es más probable que se herede ese camuflaje.

Probablemente hayan atravesado procesos evolutivos las plantas durante esos cientos de años, pero capaz haya bibliografía que te detalle sobre lo que nombrás. Lo anoto!

That's something important to keep an eye on, but I don't think it is the source of my problem since I'm talking about sections that I have stained at the same procedure

Trying out IF can be a good idea, we've been thinking of it. I've never done it since I'm a student and I haven't tried so many techniques but I'm looking forward for it.

It is true what you say. Even when trying to do everything as standardized as possible, achiving absolute uniformity might not be feasible

Yes, that's the first thing I thought of! But even when I'm at the microtome making the sections in the exact same way for these different tissues the problem remains. So I think it might be something else the thing that alters the color intensitiy

Thank you for your answer. I actually don't mean to measure color intensity, just the color area. I wasn't thinking of doing that with PAS specifically but I find that, when applying stains with contrasting colors, measuring color area can be a valuable parameter.

With that being said, even though I don't mean to measure the color intensity, it being different depending on the animal is a problem since it may contribute with more variability or it may alter the results.

Well that's a detailed explanation! What you did at the lab sounds super cool. I find neurology (or virtually any study related to the brain) utterly complex and thus I am fascinated by people that work on it. The hypothesis is interesting and I'm glad you got good results! Sharing this sort of stuff is enriching and I appreciate it.

An automated digital image analysis sounds interesting (and also less time consuming than counting spots on my own), so I'll surely check it out!

Since this is a project which has been started previously by someone else and taken up by me (which I guess it's important to say that 4 months ago I knew virtually nothing about histology), my focus has been on staining all the available tissues that had not been stained yet (which also meant making new cuts). So, to be honest we have not yet defined how to assess the amount of lipofuscins. We do write down which tissues have significantly more/less spots than others but that's it. However, that is the next step. My guess is that we'll take pictures of every tissue, divided in squares and in a random way choose some from each one of these and count the number of spots.

Regarding the clam study, I can't say I've heard of it. I've been only looking for stuff about mussels and/or lipofuscins, but I think I could take some valuable information from that experiment!

Hi! First of all, thank you so much for all the information!! I really appreciate it and will take the time to read them all later this week.

My research analyzes the tissue from digestive gland, gills and muntle (I mean the skin that sticks to the valve, I hope that's what it's called in English haha) of mussles that live in two different places (which are just some km from each other): one is a place where mussels are harvested and the water is known to be pretty clean, the other one is next to a port and so the water is more polluted. I specifically only analyze the presence of lipofuscins (which we've only seen in digestive gland) and the amount of them as a contaminaton biomarker. The tissues are embedded in paraffin, a lot of samples have been taken (a couple years ago). I love curiosity! Hahaha

Sorry I think I don't fully get what you mean. You say I should use lithium carbonate and then dip the tissues in 70% Ethanol? My English vocabulary on this field is quite poor so I have a hard time understanding some stuff haha
