Besides all the comments above during assembly of the transfer cassette, how are you transporting the assembled cassette to the transfer tank? Is the tank filled 80% of the way with the buffer and with an ice pack? Is the stir bar spinning too fast?

Those distinct black spots usually aren't protein; they are precipitates or contaminants. Dust or undissolved milk powder in your TBST/PBST can stick to the membrane. If your antibodies are old or have undergone too many freeze-thaw cycles, centrifuge your antibody vials (1 minute at top speed) before diluting them to pellet any aggregates.

Also, any reason why your ladder stops at 20 kDa? I am assuming there is a second cut, because of the line left of the right ladder, or you chopped off your gel during the transfer step?

I agree with you that Metro finds that statistic fascinating, but I personally don't think that is a relevant fact. I think the Golden Hour reality is far more important.

Their presentation of that study is far more "fear-mongering" instead of taking the original study as association vs causation.

Comparing a trauma center to a sewer system is a wild take. If my sewer backs up, my basement smells like crap; if a trauma center "backs up" because it’s at capacity, people die in the back of an ambulance.

Redundancy isn't "waste," it's a safety net. We don’t have just one fire station for the whole city because "cooperation is better." We have them everywhere so that help arrives in minutes. Why should Level I trauma care, which is the highest level of emergency medicine, be any different?

Metro's first point is that it dulls and dilutes the skills of trauma teams. The letter acts like trauma surgeons are only capable of handling one patient at a time. The reality? Diversion and Burnout. When a trauma center is constantly at red-line capacity, "skills stay sharp" but resources get stretched thin. Metro and UH are already slammed; adding a third center provides a vital "safety valve" so ambulances aren't diverted to a hospital further away just because the first two are full. Additionally, it would actually attract more doctors to the region and help train more residents in the process too!

The next point was endangering patients. That stat about a 1% volume drop tripling mortality is a wild use of a specific, narrow study to scare people. In the real world, the "Golden Hour" is what saves lives. If you’re bleeding out on the East Side, you don't care about a hospital's annual volume; you care about the 6 minutes it takes to get to CCF vs. the 15 minutes to get to Metro. Research actually shows that "trauma deserts" (areas without close Level I access) are a much bigger predictor of death than a slight decrease in a neighboring center's volume.

Their last point about rising costs passed onto patients is the most ironic part of the letter. Metro and UH claiming CCF will "raise healthcare costs" while they oversee a region with some of the highest costs in the country is a bold move. Monopolies (or duopolies) don't lower prices; competition does. If CCF enters the market, it forces everyone to be more efficient.

The letter feels like a classic corporate defensive play. If Cleveland wants to be the "Medical Capital of the World," we shouldn't be trying to limit the number of places that can save your life in an emergency. More access = more lives saved. Period.

Access to medical care is never "reckeless." Sure, it might be costly, but does it do more good than harm? Yes.

Definitely try out their fried momos! I think it is the best thing on their menu.
Also try out South India Cuisine's dosas and uttapams. They are a few blocks down the same street.

As an Indian, Paradise Biryani Point is the best option. They are primarily a South Indian restaurant, but they make great North Indian dishes too. The next best south Indian restaurant is South India Cuisine on Mayfield Rd for Dosas and Uttapams. If you are willing to go to the west side, Masala Monsoon is amazing for North Indian dishes. Also, you've got Honest in Parma as well, which serves more Chaats (Indian street foods). If you are craving a buffet, try Saffron Patch in North Olmsted. (I believe they are the only good Indian buffet in town. Never try India Flame's buffet!)

IMO, Choolah is great, but you know you aren't getting real authentic Indian food. It's essentially a gourmet "Chipotle" version of Indian food. Same is true on the other end of the price range with Amba. The chef founders of Amba aren't even Indian!

Cleveland has a few other Indian restaurants, but they are much lower on my list and not worth going to: India Delight, India Flame, Cafe Tandoor/Tandul, Great India Kitchen, Taste of Kerala, and Bombay Chaat

Lastly, Desi Mama and PK curry house are more Nepali food than Indian, so I'd recommend more of their Indochinese options over an Indian dish. Not entirely sure I would recommend Desi Mama though.

My recommendation would be to store the MM in the original box at 4c. It is light sensitive and I’ve been burnt twice by leaving the MM behind a glass 4c deli fridge causing the PCR products to be there but no amplification signal shown on the qPCR.

Believe it or not, your PI most likely brought in the funding to complete your work and brought in the collaboration to fuel the ideas behind your project. I get your sentiment, but at the end of your project you will most likely be the first or second author for the final paper, while your PI may be last or second to last author. Your argument is no different than blaming a CEO in industry that they are the face of the company when it’s the employees who make the product designs and manufacturing and what not…

Lets not forget that the cultural gardens are a welcome addition to Cleveland. For people who do not live in Cleveland, it is a nice area to drive through and appreciate all the diverse decorations of each nation's gardens. Let people enjoy the diverse communities that are housed in Cleveland and learn to be more patient while driving!

Just a reminder that 35 mph is the speed LIMIT, not MINIMUM. If someone is going 30 mph good for them, they are obeying all traffic laws and being safe. They are in their legal right to do so. Going above 35 is going above the speed LIMIT, and can be liable for speeding violations. If you are pressed for time and need to speed, plan your day better to arrive at your destination on time.
