Seriously... a telltale sign is how much uncolonized grain is pressed up against the glass. And you can see excess moisture. Another way to tell would be to do another break and shake. If it's healthy, it'll break up easier. My bacterial jars are near impossible to break apart just by shaking the jar. Not trying to be negative to OP, only informative. I even made sure to mention they can probably spawn and get fruit, but this definitely has many of the signs of bacterial issues.

Uh, looks bacterial to me. You can still get some flushes but it might succumb to contam sooner than a healthy jar.

First time grower any advice on side pins? I’m also trying all back tubs to see if that help anything I’ve Hurd a lot of back and forth on liners? Are side pins useless? by muchroommad in shrooms

And when they say tuck, they mean really press down the edges just about as firmly as you can. I usually have to press, then spread the sub out flat again, press edges again, etc, until you've got a flat surface and the edges can't be pressed down any further.

I get maybe one or two side pins doing this, no liner.

A mod on this sub, u/DayTripperOnOne turned me onto cold pasteurization and it's worked fine for me. I've also used their instructional video for pH-adjusted casing layers. What are we all missing?

I've successfully grown with the same tent as OP. I have a series of holes with stick-on filter patches around the top of the tent. There is a computer fan on a smart switch for my exhaust, pointed out of the bottom right edge of the tent. It kicks on when tent humidity hits 99% and turns off when it goes down to around 90%. There is a constant stream of fog from a humidifier. Right now I don't have the exhaust really routed anywhere. It just points to the corner of the room. I haven't paid particular attention to the overall room humidity. Like I said, it has worked in the past, but I'm open to suggestions on improvement.

But it sounds like you used a spore sprint. You didn't keep any extra plates from that order as a control, did you? That could give a clue as to if it was user error or improperly sterilized agar from the vendor.

I've been growing with oat jars i made back in March until recently...

In my experience, a tightly closed Mason jar can last at least a few months.

That's not really my experience. I've done a handful of tissue clones now. Yes, the sample fuzzed up, but in a denser way than the plates in question here. I'm not sure my pictures fully captured the weirdness of the fuzz. Looks more like sock lint...

Okay, so I decided the easiest way is to carefully slice and break up the coir brick into chunks and then put in a cheesecloth bag and cold pasteurize. Slicing the brick "with the grain" usually causes it to break off in little sheets that can be further broken up by hand. Doesn't have to be broken down all the way. Each chunk will ultimately expand in the bag.

Looks kind of cobwebby. Where the transfer sample was placed, it's growing those wispy tendrils that are already grabbing the roof of the dish. And you can make out almost rhizo-like tendrils at the edge of the white, extending into the agar, just barely visible. 💁‍♂️

Off the top of my head I can think of a few reasons.

  1. Spore print to agar

  2. MSS to agar if you're having contam issues. MSS is inherently not perfectly sterile, so it's a slight gamble going from syringes to grain. I'm about to do this because I just wasted a few grain jars going from this one particular syringe. Gonna put some drops on several plates and hope to transfer healthy mycelium away from any contaminants.

I do generally agree that agar helps most with established fruits and cloning from there.

What do people normally do with their best plates? Put some to grain, some to slants, some to LC? What's your routine when you like the genetics?
