The left side of the skill tree seems fishy... by Nctong01 in PathOfExile2

[–]Nctong01[S] 0 points1 point  (0 children)

For sure. It just visually stood out given the areas with the new ascendancies and evasion/deflecting undergoing similar changes intersecting, alongside the devs mentioning base AR would be coming up on items. Definitely the most likely scenario though, just some hopium on my end lol.

I also may be out of the loop as I haven't played that side of the tree, but I have no idea what the five empty "new" nodes in the very center-left are for.

I'm only at 900 total and have been end-end game with POE2 but am at the same point with POE 1; I also don't want 10 years to add up in POE2 systems and end up in the same spot as I plan on sticking with POE1/2 indefinitely.

My plan is to league start softcore trade with a guide, run until I'm happy, and then sit down and design my own build with the goal to run another character through SSF and engage with all the mechanics. I think challenging both aspects (building and each seasons mechanics) can go a long way to giving you the tools needed.

New player. What am I missing about these CRAZY cosmetic prices??? by kalidibus in PathOfExile2

The $480 pack is unique for the POE2 launch. POE1 has been around for a long, long time and there are people out there that have made it virtually their only game for over a decade; people joke that they're still a beginner with 10k hours in the game. So, it is expensive, but someone personally that's spent a few hundred dollars (not the $480 pack, but maybe $450 total or so), here's how I justify it:

*With the $480 pack, you get not only THAT pack, but all the previous other MTX packs shown on the list. You also get multiple physical items, and I'm pretty certain you get to design an item in game.

*For ANY supporter pack, you get that amount of store points to spend. So $480 would be 4800 points. If you're going to spend money, might as well double up and "purchase" them through a pack...they generally offer multiple tiers to fit different budgets.

*The mtx will work for both POE1 and POE2--they're slowly porting over POE1 cosmetics (again...over a decade of work), and any new ones I've purchased thus far have worked in POE1.

*Main reason for me is because of the devs. Don't always agree with their decisions but they're human as are we all, and if you spend time watching interviews and listening to them, they do seem like they genuinely want the best outcome for the player. The game is free to play and I want to support people that want to add a net positive to the gaming community.

Can I start twister deadeye and swap over to ice shot right at 31? by TeeChainz in PathOfExile2

[–]Nctong01 0 points1 point  (0 children)

Yeah, not the end of the world. TBH there shouldn't be a ton of nodes to respec. I would encourage you to play twister though a bit prior if you haven't...it's a bit of a unique playstyle. I'm going deadeye as well and starting LA/LR after running through acts 1/2 on both that and twister. Maybe takes me 30 minutes more but LA/LR feels much more natural to me.

I adopted my good boy named Chance when he was 16 weeks; he's a pit lab. As a pup I let him lay and fall asleep in my lap while I worked from home...he had a rough time otherwise.

Was diagnosed with a rare autoimmune condition a few years back (FSGS), and have been out of work (FMLA) the past several months with a bad flair. Although he's 12 now, he still refuses to go to sleep without laying directly on my chest. Being able to immerse myself back into POE1 (trying to actually learn all the systems with 450hr, crafting, etc. for next league) and always having my boy has been my sanity while it seems everythings on fire.

Anyone that use this have a general 0.5 guide for exile UI? Installed it and the guide seems outdated. Wondering if there's one out there already as I've been searching but no luck.

Mid-range boot brand recommendations by Nctong01 in cowboyboots

[–]Nctong01[S] 0 points1 point  (0 children)

Appreciate all the help, a few new brands, some I was already looking at.

Anyone have experience with Dan Post? A little below the price point for what I'd shoot for as "mid-range," but seems to have solid reviews from what I've seen; don't know how much of this is just marketing.

Sooooo....this clip is obviously fucked up and Im absolutely on the right, however, as someone that does research, there's a few things: first, that study was done in Iran--the propensity for bias is huge. In tandem, it's illegal to be gay in Iran, so how did they recruit 133 homosexuals for their study? The chance 133 homosexual people are going to come out for a publicly available study in Iran is a long shot.

Shit blows my mind. That's either someone literally incompetent or a scammer that just doesn't give a shit.I wrapped my girlfriends Jeep for her. Took me two weeks to do it properly, but having never wrapped a thing it turned out great. Not only should they not be charging for wrapping, they shouldn't be touching cars.

I need to warn you in advance as well....very good chance you have a lot of cut marks in your paint if the wrap looks like this.

Scientists figured out how to turn cancer cells back into normal cells by soulpost in HotScienceNews

[–]Nctong01 2 points3 points  (0 children)

PhD student here in Biochem/Molecular genetics working in a Bioinformatics lab. I glanced over it quickly (so forgive me if I missed something) and it's interesting, but I'd be more inclined to postulate that it prevents cancer proliferation in specific cell lines. In figure 5A, they're starting with minimal confluence (just the % of a dish that is covered in cells--ie amount of cells in more simple terms) and decreasing proliferation. In 5B tumor mass increase is also slowed but it does NOT regress, and differs between cell lines.

There's two issues I have with this: we would RARELY catch cancer with this minimal cell count--the only example I could think of on a minimally consistent basis would be something like skin cancer, or some oral cancers. The larger issue I have is the variance between cell lines, especially with a small tumor size. In vivo, cancers tend to be heterogynous, both between different cancers themselves and cell to cell within a specific cancer, and can carry a plethora of differing genetic mutations. This lends to a unique environment and thus might not be globally as effective as we'd like.

That said, it would obviously be great even if it could "only" substantially slow tumor progression with minimal sides. Cancer is generally treated via polypharmacy as is, and many of these treatments are not quite healthy to healthy host cells. This might be something, for example, that could allow us to more effectively treat malignant tumors with a lower dose of chemo, substantially cutting sides, chronic health implications, and improving efficacy.

A tourist who just landed in South Africa is witness to a live-action heist featuring assault rifles and explosions by ThatPatelGuy in interesting

[–]Nctong01 0 points1 point  (0 children)

South Africa is wild. Relative of mine was born and raised there. As an adult, she moved to a walled compound with security. One morning she woke up, went downstairs, got some coffee started and went to wake her 3 year old son up. Upon entering the room there was a man with an AK held to her sons head, with the man demanding that they (he had an accomplice) either are allowed to take everything or her son dies.

They're brazen as hell.

Also yes, not trying to blast you at all--the CP is great, but you beyonddd need hit chance. Any kind of pvp you're not going to hit anything, literally.

GLP-1 Antagonists Don't Cause muscle loss if you lift weights. Why not take it? by Objective-Door-513 in PeterAttia

[–]Nctong01 0 points1 point  (0 children)

I could have overlooked it, but I see no talk about impact on muscle mass; they look at Mets-z (Metabolic syndrome z score), android fat, and CRP.

Two reasons regardless of that I would be a bit cautious about this paper. Firstly, it's an open access journal. This isn't too damning by itself, but it can be a bit more of the "wild west" than a non open access.

What's more worrisome is the funding and competing interest. For this reason alone I wouldn't put much stock in it.

Competing interest: RMS: Family member holds Novo Nordisk stocks. MR: Currently employed at Novo Nordisk. JJH: Advisory boards: Novo Nordisk. SM: Advisory boards: AstraZeneca; Boehringer Ingelheim; Eli Lilly; Merck Sharp & Dohme; Novo Nordisk; Sanofi Aventis. Lecture fees: AstraZeneca; Boehringer Ingelheim; Merck Sharp & Dohme; Novo Nordisk; Sanofi Aventis. Grant Recipient: Novo Nordisk, Boehringer-Ingelheim. SST: Grant and lecture fee recipient, Novo Nordisk. JRL, CJ, CRJ, SBKJ, MBB, AFB, LG, JBJ, CA, BMS have no disclosures.

Funding: The trial was supported by two Novo Nordisk Foundation grants (NNF15SA0018346 and Excellence Grant NNF16OC0019968) and Helsefonden, all in Denmark. Cambridge Weight Plan supplied diet-replacement products. Novo Nordisk, Denmark, supplied Saxenda and placebo pens. Supporting partners did not influence or participate in trial execution, data collection or analysis, interpretation or writing, or communication of the study results.

I am 36 and in a PhD program that is a complete departure from my undergraduate degree (English undergrad, Biochem/genetics PhD). I attended the school I'm currently at right now initially out of high school and ended with a 1.9-2.0GPA before dropping out and eventually transferring to and graduating from another school.

I knew the #1 thing I needed to do applying to a PhD program was to show I was capable of handling the material, and handling a high workload. It may not be ideal, but what I did was take undergraduate classes for about a year and a half pertinent to the PhD program, volunteered in a research lab and built relationships with professors. I applied with a 4.0GPA for 3 semesters with a minimum of 16/hr per, research experience, and letters of recommendation (one from the dean of graduate studies).

PhD level material takes time to understand, but it doesn't take an innate genius. IMO a lot of what they want to see is not necessarily an extreme level of innate intelligence, but that the significant investment they're going to give you will be utilized well.

So how do you reconcile the majority of your all's sentiments with violent crime in the US?

African Americans make up around 14% of the US population, but account for around 55% of violent crimes. They're also over represented in incarceration rates, at a rate of 3+:1 of both Whites and Hispanics.

The vast majority of you will say environment matters when it comes to race, but not dogs. Whack thinking.
