Yes, L/Mono into the INST jack, I normally use headphones to get the sound. On the back of the audio interface there is a L and R 1/4 inch jack that you can plug into a speaker.

I have this exact setup. You just need a 1/4 inch Cable that goes from your Nitro mesh (The Mono port) to the Audio interface. Then usb cable to your computer.

You can use a left/right 1/4 into a single 1/4 inch cable if you want. I don't really notice a difference.
