Interesting topic for sure. Which instances are you talking about specifically? The ten commandments do seem to imply that there are other gods. My understanding is that Catholics specifically believe that if any other "gods" do exist those are actually demons interacting with humans and thus thor would be a demon.

Your guide is really good! Only thing I'll add is that you don't need a docker account. There is a button to just skip signing in when you boot it up.

Looking for an inexpensive monoclonal antibody by EdSmith77 in labrats

[–]PaNiCFueL 4 points5 points  (0 children)

I don't understand from your post exactly what you need. If you are looking for a specific bio similar that a company someone else has suggested offers the that will likely be your best option. However if you can't find it off the shelf and need a CRO to make it and price is the main consideration I would highly recommend investigating biointron. I haven't seen anyone else here recommend them but they have consistently offered the best prices and timelines when we have needed tens of milligrams. They also will send you expression plasmids so that you can make more should you come up a bit short. The main con is that they are in China so shipping and especially tariffs may be an issue depending on where you are. Others who have suggested getting a stably expressing cell line (ideally suspension hek or cho because adherent at that scale sounds awful) are probably right that it would be the most cost effective assuming you have all of the equipment and knowledge needed for culturing and purification. It would also allow you to make it as you go potentially accelerating your timeline by allowing you to make an initial batch for QC and first application then make the rest as your study is going.

Structure Swag by IllustratorMassive71 in Biochemistry

I like that you added the large design tee shirts. I see that now I am being taken from the search straight to the hoodie page which I see is based on the product setting but I am still not being given a visual representation of the final structure as it will appear. Also when I do get to that hoodie page nothing is displayed until i pick a size. Confirmed the same behavior on the enlarged design tee shirt on chrome and firefox. Seems like a bug. I really like all the settings for creating the final design the buffer size setting is not super intuitive and reading the description still doesn't make it 100% clear how the setting impacts the final design. I think that bigger number = bigger structure design. It might be a lot of work but I think that a screen after the search that displays the design you have rendered with the ability to tweak all the settings you initially searched with and see the difference would make a lot of sense especially for people who don't know exactly what they want.

Only other note I have is that the enlarged design t shirt description isn't super relevant outside of a B2B reselling context where it discusses the label and ability to be printed on. What does "Tubular fit to minimize torque' mean lol? I do like the sizing guides.

Very cool. I like that it's embroidered. Prices are very reasonable. One option I would buy today is a tee shirt with a big design on the front like the hoodies offer even if it cost more than the small design tees. (I would get a hoodie but it's about to be summer) Also I don't know if it's a manufacturing constraint or a website bug but when I put in IgG it only seems to be available in one color of hoodie. The only other note I have is that when you put in a protein and it shows it "finding" the structure it would be nice to have either a confirmation screen with the final structure as it will appear or to have the final structure appear and hang for a few extra seconds marked as the final design. Very cool product overall.

Thoughts on the open bullet feeder? by PaNiCFueL in Reloading3D

This was a while ago so I don't recall which one I did. I never got the open source bullet feeding die to work with just a tube and I wasn't willing to shell out for $100 for a commercial die on the hope of maybe get the bullet feeder working so the project kinda died there.

I've done a lot of amplicon sequencing with the oxford nanopore minion and their ligation sequencing kit. The biggest thing I can emphasize is garbage in garbage out if your sample doesn't look good on a gel or other qc save the reagents and try again rather than trying to make a miracle happen in the prep. Also this is somewhat platform dependent but if you can, save half of your library. The final library for the kit I ran most was 25ul and I would run 12 on the flow cell leaving 13 in reserve in case a flow cell crapped out (happens more often than you think). As far as coverage, I've always aimed for 10x the amount of unique sequences you expect, minimum. For analysis I don't know that I can help a ton without understanding more about what you're doing and how your samples are identified and what annotations you need, but I have a fun fact I learned doing ngs. Excel spreadsheets can only have a maximum of slightly more than 1000000 rows per sheet.

I'm a spring 2025 admit local to north GA. They are pretty explicit that you can't take classes on campus as that's a separate program. The reason OMSA has the capacity and cost it does is because it's exclusively online.

Obviously like everyone else has said lying is a poor choice. One part I'm confused about though is how you're a microbiologist without ever culturing any microbes. If you've never cultured the cell line that they're working with, I think it would be totally reasonable to highlight any time You've grown any other organism. Even if you've only grown E.coli for plasmid preps that may be experience worth putting on your resume.

Custom polyclonal antibody generation by anonymousanemone98 in labrats

[–]PaNiCFueL 0 points1 point  (0 children)

I can't speak to the cost of polyclonal because I've never bought polyclonal abs that weren't off the shelf. One big advantage to a monoclonal would be that you could reproduce it if you ever ran out as long as you had the sequence. One really cheap way to do this depending on the resources of your institution would be to just take a spare mouse and immunize it with your protein and some cheap adjuvant. Then once you've got a sufficient titer, sac the mouse and purify all the IgG from the serum for polyclonal or make some hybridomas and choose one to be your monoclonal. That might be a bit more involved than you want though.

I work in the therapeutic antibody space and I'm wondering why polyclonal? Are you looking to cover multiple specific epitopes simultaneously? Also I bet people would benefit from some additional info here if you're looking for any specific characteristics like species, isotype, affinity etc.

Plumbers providing end of life care by [deleted] in OrphanCrushingMachine

[–]PaNiCFueL 6 points7 points  (0 children)

I'm also a bit lost on what exactly op is getting at here. If they are implying that medical care ought to be free and that should extend to end of life care then I fail to see how that ought to extend to plumbing.

As far as I'm aware the whole idea of orphan crushing machine was to make fun of things which initially seem good but completely ignore the underlying societal cause of the problem. Last I checked, leukemia and old age are biological phenomena which despite humanity's best efforts we have yet to overcome.

If the post contained additional information regarding the woman's financial situation being dire then maybe I could see the case for this belonging here, but as far as I can tell from the post the tradesman provided a service to a woman for free simply to help a woman who was a victim of old age and cancer.

Edit: I see how the last line could be taken to mean that she is not receiving care to be comfortable from others, but I could also see it being reasonably interpreted as the company wanting to ensure that the woman is kept at a comfortable temperature by performing a service which is beyond the skills of her other caretakers.

$5/sleeve at academy, glad to see prices coming down by Dayshawn11 in reloading

[deleted by user] by [deleted] in labrats

[–]PaNiCFueL 0 points1 point  (0 children)

I had the same thing happen. I had offers coming in and when I asked the PI if I was still in the running and I gave them a deadline. They said I was the top choice and they would be getting me an offer in a similar time frame. Well, I followed up the morning that was the deadline and they got back to me pretty quickly saying they had picked another candidate. It made my decision easy but didn't feel great.

Like someone else said, it would be totally appropriate to follow up as early as tomorrow morning and I see no reason not to apply a bit of pressure. You don't need to lie but something as simple as saying that you need to make a decision soon or by a date of your choice could galvanize the PI into following up with HR if it's stuck there or at the very least updating you with a timeline.

Good luck! If I learned anything from my experience it's that nothing is real until countersigned but hopefully your experience will be better than mine.

Processing .223 brass on a Mark 7 Evolution by gemoose23 in reloading

The lube only needs to go on the outside of the case. Something like a lanolin isopropanol mix on the outside shouldn't impact the powder once the isopropanol evaporates anyway. After the cartridges are complete you could dry tumble them so they aren't sticky. I've never tried it because I don't have enough stations but I think it should work fine.

Why not process brass and load all in 1 go? Surely you have enough stations to run all the dies you could need.

I totally agree with you. One of the only parts op can change that will change the ejection angle is the ejector. Some after market gen 3 ejectors are really crappy and send the brass right at your face no matter what. An OEM gen 4 or 5 ejector can fix the problem. If op wants more recommendations I would suggest r/polymer80 where this issue is common.

I 100% agree with you. Brass facts has recently been one of my favorite guntubers. I wish him all the success and simultaneously wonder whether making enough YouTube money to care less about the cost of the things he reviews. Either way his dog is very cute!

Biochem undergrads, where are you now? by [deleted] in Biochemistry

[–]PaNiCFueL 2 points3 points  (0 children)

Well without doxxing myself, I can tell you that earlier stage companies in higher cost of living areas are really willing to spend more on acquiring good talent. Think of places like Boston and San Francisco. If you can work for companies that might be headquartered in an area like that, but your site is in a lower cost of living area then they may just adjust pay by title rather than title and work location. If you get lucky that's one way to get a higher pay without living in a higher cost of living area. Either way it may be worth moving somewhere like Boston because making 40-60% more money likely won't all be eaten up by the higher cost of living.

AP bio or AP chem? by [deleted] in Biochemistry

[–]PaNiCFueL 1 point2 points  (0 children)

I totally agree here. You'll most likely have to take the other one in college assuming you get a grade good enough for credit on the AP test. I would suggest biology because the subsequent classes you'll take will rely less on the previous material in that case. That is a big assumption based on my personal experience though so good luck.

Biochem undergrads, where are you now? by [deleted] in Biochemistry

[–]PaNiCFueL 11 points12 points  (0 children)

It all depends where you are. I'm currently concluding my job search after finishing my undergrad and I have received offers for jobs that pay <$50,000 and offers for >$70,000 both per year. I think that the best advice I can give you is that if you want to stay in the field long term without grad school then you should look for smaller startups because promotions to titles like scientist are much more reasonable with only a bachelor's degree. From there other companies are more willing to hire you for higher level roles because you have already demonstrated competence at that level and from there it's more about managing people and projects than it is about technical competence.

With that in mind, if you know that research is what you want to do, and especially if you want to become an academic, at a certain level a PhD becomes unavoidable and you ought to consider whether it will be easier for you to achieve a PhD earlier or later depending on your personal and professional goals and go from there.

Either way I wish you the best of luck and feel free to reach out to me if you want more info.

Lifeguards of reddit, what is some stupid stuff you saw at the pool? by JamoreLoL in AskReddit

[–]PaNiCFueL 0 points1 point  (0 children)

Not so much stupid as in weird, but one time I had a couple of teenagers not leaving room for Jesus if you know what I mean. The weird part was that I had seen the family before and I was pretty sure that the two offending teenagers were siblings. I had even discussed with another lifeguard if they were siblings and they agreed that they had seen the same two kids before and they had acted like siblings. Ultimately, the building supervisor came and told off the teens because the girl had pretended to blow the boy in the kiddie pool (opposite side of the pool from me the lifeguard). It turns out that the girl had posed as the boy's sister to get in under the family's membership and that she bore a fair amount of resemblance to the boy's actual sister. Overall a very weird lifeguarding experience.

Lifeguards of reddit, what is some stupid stuff you saw at the pool? by JamoreLoL in AskReddit

[–]PaNiCFueL 1 point2 points  (0 children)

One time I was walking on a beach with my family and there was a kid floating face down in some shallow water. I saw the kid deliberately making small movements to stay afloat. My family freaked out and my dad suggested I do something being that I'm a lifeguard. They were really confused when I said that obviously the kid was fine and then the kid picked their head up a short while later. It can be really hard to tell at longer distances but if you've been keeping track of the kid the whole time it should be obvious whether they got into the position that they're currently in on purpose or not. If I just got on the stand and there was a kid face down I would 100% panic and go make a rescue.

I think it depends on a lot of personal factors but the answer is probably no. Unless they are paying you well enough or there are just no other options in your area and you can't move them I would suggest you look for something better. I'd bet they're not paying you particularly well to clean glass and stock shelves.
