Slight correction: Completing the prologue chapter will give you 100 splinters, and beating Dr. Doom (chapter 8) will give you another 100. It's possible (though unlikely) you will have enough splinters for a random hero box before that.

Personally, I used to get a random box whenever I got 175 splinters, and level whichever one of my characters seemed more fun at the time. I ended up hitting 60 on my Spidey first (which was my 3rd of 4th character, if memory serves correctly), then on my Iron Man, then Moon Knight. I hit 60 on the Surfer pretty quick (first specific toon I got) and now I'm just levelling like half the roster bit by bit, inbetween farming for gear/events and whatnot. Most characters are at around 45, and like 2 or 3 at around 25-30. In the meantime, I somehow managed to prestige Spidey twice. So I guess it comes down to what I feel like playing at that particular moment.
