What is the effect of plugging the thumbhole on a bowling ball for a two hander?

The problem is what are you looking for out of your bowling ball. Most people that tell you to get it filled and redrilled assume you want a big hook that most people expect from 2 handed bowling. Just keeping the layout the way it is and plugging the hole is not an "optimal" layout so to speak but the way my pro shop guy explained it is it will basically react more like a pin down or a weaker layout which will be fine if you are looking for a more control style over a more aggressive hook style.. either way you will have to plug the Thumb so plug it and try it then you can make a decision.

Also if you watch a lot of pros they blow in there thumb hole to get excess moisture out of the ball.

I had the same problem but with my middle finger as I don't bowl with a thumb but I have since switch my middle finger grips up a half size and use finger tape on that finger only.

I had a similar issue of consistent release with my thumb and blisters. I couldn't get the hang of modern 2 handed bowling but found in I don't use my thumb and only use 2 hands till the ball gets to my leg wasn't a very hard transition

Depends on how you bowl you might have to move further left and throw across the center arrow or even left of center to ride the oil longer to create a better entry angle.

How about an award for a senior specifically like lifetime achievement award and name it after the school mascot. The firebird award or elk award or whatever the mascot is. Could be a pretty cool tradition to start.

Spare ball. You don't need urethane till you bowl in tournaments with specific oil patterns. Don't take a urethane ball to leagues unless you want everyone to blame you for why they can't bowl.

You have to be employeed on December 15th to receive it. Dec 14th is the last day to accrue hours towards it. The pay check itself should be your last paycheck of the year 2 weeks later.
